低温胁迫是影响水稻产量及品质主要因素之一。发掘水稻冷胁迫调控蛋白及对其耐冷的分子机制研究对耐冷水稻分子育种有重要的理论和生产实践意义。前期利用iTRAQ标记定量蛋白组学方法鉴定到耐冷水稻空育131中1个冷胁迫响应蛋白CORP1在低温处理24h、48h和72h正调控幅度较大,且与转录水平调控趋势一致;验证了日本晴的CORP1突变体耐冷强度降低。本项目拟进一步从蛋白表达水平验证CORP1在低温胁迫下的调控模式,并通过基因定点敲除、过表达基因功能互补实验,明确CORP1参与水稻耐冷的分子功能;利用YFP荧光蛋白融合表达技术明确CORP1在水稻中的亚细胞定位,并利用免疫共沉淀技术探索其相互作用蛋白,通过进一步分析这些蛋白在低温胁迫处理水稻中的表达模式等明确CORP1调控水稻耐冷的分子机理。预期研究结果将基本阐明CORP1蛋白调控水稻耐冷的生物学新功能及其分子机制,为耐冷水稻品种分子育种提供新思路。
英文摘要
Low temperature stress is one of major factors which influence rice yield and quality. Discovering rice cold stress regulation proteins and studying the cold tolerance molecular mechanism of these proteins have important theoretical and production practical significance for molecular breeding of cold tolerance rice cultivars. In previous study, a cold stress response protein CORP1 was highly up-regulation at low temperature treatment for 24h, 48h and 72h in cold tolerance rice cultivar Kongyu131 was validated by iTRAQ labeling quantitative proteomics approach, and the trend of protein regulation was similar to the transcriptional level regulation; the cold resistant level of Nipponbare CORP1 mutant decreased in the validation experiment. In this study, we will further validate the CORP1 regulation patterns of protein expression level under low temperature stress, and reveal molecular function of the CORP1 that involved in rice cold tolerance by targeted knockout gene, function complement experiment with gene over-expression. The CORP1 protein subcellular localization will be validated using fluorescence protein YFP fusion expression technology. The rice cold tolerance molecular mechanism regulated by CORP1 will be revealed by exploring the interaction proteins with co-immunoprecipitation approach and further analyzing the expression patterns of these proteins during low temperature treatment in rice plants. The prediction research results will basically illustrate the new biological function and molecular mechanism of the CORP1 protein which regulate rice cold tolerance, and provide new ideas for molecular breeding of the cold tolerance rice cultivars.
