塑料已经成为现代社会不可或缺的产品而被广泛应用,随之而来的微塑料污染也成为全球性的环境问题,微塑料的环境安全性研究是目前塑料制品发展亟待解决的关键科学问题。基质辅助激光解吸电离飞行时间质谱成像技术(MALDI-TOF-MSI)是一种结合质谱分析和影像可视化的新型分子成像技术,具有原位、可视化、多目标分析等优点。本项目围绕微塑料对水生生物的毒性效应及机制的重要科学问题,以MALDI-TOF-MSI为主要手段,拟开展微塑料在生物体的生物过程及致毒机理的研究,研究微塑料在斑马鱼体内的吸收、运转、分布和代谢以及毒性效应,阐明微塑料在斑马鱼体内的作用过程,明晰微塑料对斑马鱼的毒性效应,探索微塑料暴露的分子标志物,构建毒性效应与作用过程的关系,揭示对斑马鱼的毒性效应机制,该研究对阐明微塑料的环境健康与安全性,建立微塑料的环境质量标准和拓展微塑料的原位检测方法,具有重要科学意义。
英文摘要
Plastic has been widely used in modern society, and microplastic pollution has also become a global environmental problem. Study on environmental safety of microplastics is a key scientific problem to be urgently solved in the development of plastic products now. Matrix-assisted laser desorption/ionization-time of flight mass spectrometry imaging (MALDI-TOF-MSI) is a novel molecular imaging technique combing mass spectrometry and image visualization, which has the advantages of in-situ, visualization, multi-objective analysis and so on. This proposal will focus on the biological process and toxic mechanism of microplastics in organisms by use of MALDI-TOF-MSI. The main objectives of this proposal are to (1) develop the analytical methods of microplastics; (2) learn about the absorption, transport, distribution, metabolism and toxic effects of microplastics in zebrafish; (3) elucidate the action process of microplastics in zebrafish; (4) clarify the toxic effect of microplastics on zebrafish; (5) search new biomarker for exposure of microplastics; (6) construct the relationship between toxic effect and action process; (6) reveal the mechanism of toxic effect on zebrafish. This study is of great importance to clarify the environmental health and safety of microplastics, to establish the environmental quality standard of microplastics.
