神经系统疾病给社会和个人带来沉重负担,而甲基汞是一种能导致神经系统疾病的环境污染物。肠道菌群在机体中扮演重要的角色,但它们对甲基汞神经毒性的调控作用仍不清楚。本项目将综合利用高分辨同步辐射技术,如SR-XRF,SR-PCDI及SR-XAS以及金属组学、宏基因组学、代谢组学等手段,首先以肠道模拟系统为模型,探讨肠道菌群对甲基汞的吸收与转化及甲基汞对肠道菌群结构及代谢产物的影响情况,然后以大鼠为模型,确认体外研究结果并探讨甲基汞如何穿过胃肠壁及血脑屏障,最后探讨益生菌摄入或肠道菌群移植对甲基汞神经毒性的调控作用。通过综合研究肠道菌群对甲基汞的吸收与转化情况,甲基汞对肠道菌群结构及其代谢产物影响情况,益生菌摄入或肠道菌群移植对肠道菌群结构及与肠-脑轴相关的代谢产物的恢复情况,阐明肠道菌群调控甲基汞神经毒性的作用机制,为提高甲基汞暴露人群健康水平探索新途径,为帕金森病等病的预防和治疗提供有益借鉴。
英文摘要
Neurological diseases bring heavy burden to the society. Methylmercury (MeHg) is an environmental pollutant that leads to severe neurological toxicity. Gut microbiota have been found to play vital roles in life processes, however, their roles in modulating the neurotoxicity of methylmercury are not clear. This project will work on this using in vitro and in vivo models through synchrotron radiation based high resolution techniques like SR-XRF, SR-PCDI and SR-XAS and metallomics, metagenomics and metabolomics methods. We will first study the absorption and transformation of MeHg by gut microbiota and the perturbance of the gut microbiome and its metabolic profile especially gut-brain axis related metabolites by MeHg using simulated intestinal microbiota ecosystem. Then we will check the results got from in vitro study and investigate how MeHg cross the gastrointestinal wall and blood-brain barriers in rats. Finally we will study the effects of probiotics intake or fecal microbiota transplantation on the modulation of neurotoxicity of MeHg in rats. In combination of all these findings, we are hoping to elucidate the mechanism underlying the modulation of neurotoxicity of MeHg by gut microbiota, which will offer new alternatives on health promotion in mercury-exposed populations. Our study may also shed light on the prevention and treatment of other neurological diseases like Parkinson’s disease and Alzheimer’s disease.
