麻痹性贝毒是由甲藻产生的全球近海危害最严重的藻毒素之一,在我国近海危害也十分严重,中毒事件时有发生。目前在全球近海已报道的麻痹性贝毒组分有54种,在我国近海报道的有23种(类),我国对麻痹性贝毒新组分缺乏系统研究。本研究通过批量培养有毒微藻、诱导微藻产毒,典型海域采样、现场富集毒素等手段,获得足够量的染毒实验材料。先用亲水色谱柱-液相色谱串联质谱法检测技术,发现我国近海微藻及海水和动物体中麻痹性贝毒新组分;再用大孔吸附树脂和质谱指导制备型高效液相色谱相结合的方法获得毫克级高纯度(≥99%)毒素新组分;最后用液相色谱-高分辨质谱和核磁共振方法确定分子结构及纯度。采用细胞毒性试验和小鼠生物法确定新组分毒性。系统阐明我国近海麻痹性贝毒新组分及其毒性,对科学评估麻痹性贝毒危害,具有重要意义;可制备出麻痹性贝毒新组分参考物质,对加强我国近海麻痹性贝毒监测,保证海产品食用安全,具有切实的应用价值。

Paralytic Shellfish Poisoning (PSP) is produced by the dinoflagellates in the ocean and has been one of the most harmful algal toxins in the world, and the harm is also very serious in the coastal waters of China. There are only 23 kind of PSP components has been reported in the coastal waters of China, and the more than 54 kind of chemical components had been found in the world. There is no systematic study of the profile of toxin new components in the coastal water of China. In this study, the large scale culturing of PSP-produced microalgae and inducing the algae increased toxins secretion in the laboratory will be used to obtain the sufficient toxic material for finding the new PSP components. To determine the new toxin structure, the microalgae and seawater, typical susceptible fishes, shellfish, crabs, sea urchin and stars which may accumulate toxins, will be collected in the 6 typical stations in the coastal waters of China. Firstly, the technique of HILIC-HPLC-MS/MS will be used to find the new PST components in the samples. Secondly, macroporous adsorption resin and preparative high performance liquid chromatography will be used to obtain high purity(≥99%) toxins. Lastly, the techniques of HRMS and NMR will be used to identify the component molecular structure and purity. The cytotoxicity assay and mouse biological method will be used to determined the toxicity of new PST components. The new PSP composition and toxicity in the coastal water will be clarified systematically, and will scientifically explain the PSP harm in the coastal waters of China. This study is therefore critical to prepare the PSP standard reference materials to strengthen the toxin monitoring and ensure the safety of seafood consumption and human health in the coastal water of china.
