植物株型是一个重要的农艺性状,与植物的成熟期、机械收割以及产量息息相关,而植物株型很大程度是由叶和分枝的发育来决定的。已有研究表明植物的分枝发育是一系列转录因子和不同植物激素协同作用的结果,但其精细调控网络还需进一步的深入研究。近期研究发现,拟南芥法尼基转移酶突变体era1-2在分枝发育方面存在严重缺陷,表明法尼基化修饰参与了植物分枝的形成过程。为了进一步探讨法尼基化对植物分枝发育的影响,本研究拟以era1-2突变体为材料,1) 分析不同转录因子和植物激素与法尼基转移酶在植物侧生分枝发生过程中的相互关系,2) 通过蛋白质组学和EMS诱变筛选era1-2突变体抑制子,寻找法尼基化修饰的底物,深入挖掘相关底物对植物分枝的调控作用。本研究将进一步揭示法尼基化修饰调控植物分枝的分子机理,提高对蛋白翻译后修饰与植物株型之间互作网络系统的认识,具有重要的理论意义和育种应用价值。
英文摘要
Plant shoot architecture is an important agronomic trait, which strongly influences plant ripening, mechanical harvesting and yield. And the shoot architecture is largely determined by leaf and shoot branching development. Previous studies have shown that shoot branching is regulated by several transcription factors and different hormones, but its fine control network needs further research. Recent work indicated that mutation in the protein farnesyltransferase (FTase) era1-2 led to strong branching defects in Arabidopsis, suggesting that protein farnesylation modification was involved in modulating the shoot branching development. To investigate the role of farnesylation in shoot branching, we would like to analyze era1-2 mutant, and focus on the crosstalk between farnesylation and different transcription factors and hormones. In addition, we would find the targets of farnesylation through proteomics and EMS mutagenesis. In conclusion, the work will uncover the molecular mechanisms of plant architecture underlying the farnesyled modification, and provide a new hint between plant architecture and protein post-translational modification, which will help yield improvement in crops.
