骨关节炎(Osteoarthritis,OA)属中医痹证。研究表明外界温度可刺激Thermo-TRPs/HSPs通路调控OA,这暗合风寒湿致痹的中医理论,且长久以来用桂枝汤或其变方为代表的温通中药治疗痹症均有显著临床效果。基于前期研究,发现风寒湿可影响OA结构改变,且桂枝汤的靶点与前述通路有关联。从而提出假说:风寒湿介导Thermo-TRPs/HSPs交互串话引发OA,温通中药可对二者良性调节从而延缓OA。.项目在前期工作基础上,拟从现有FHS队列中采样,前瞻追踪2年,探讨风寒湿影响Thermo-TRPs/HSPs与关节结构性变化的因果关系;再用风寒湿状态的大鼠模型,观察风寒湿影响此通路的状态及桂枝汤的作用靶点;再以风寒湿模型大鼠血清干预软骨细胞,探究风寒湿介导Thermo-TRPs/HSPs的串话特点及桂枝汤的干预机制。研究有助揭示中医经典理论的物质基础及经方中药治疗OA的科学价值。

Osteoarthritis (OA) is Bi syndrome of traditional Chinese medicine. Studies have revealed that external temperature can stimulate Thermo-TRPs/HSPs pathway, and it is relative to the regulation of OA. This mechanism is consistent with the theory of traditional Chinese medicine, which is called wind-cold-dampness inducing Bi. Besides that, for a long time, Wentong traditional Chinese medicine represented by Guizhi decoction or its variants has achieved remarkable clinical effects in the treatment of OA. Based on previous studies, it was found that wind-cold-dampness could affect the structural changes of OA patients’ knees, and the target of Guizhi decoction was related to the Thermo-TRPs/HSPs pathway. Based on previous studies, it is hypothesized that wind-cold-dampness may induce OA by mediating Thermo-TRPs/HSPs interactive crosstalk, while Wentong traditional Chinese medicine can regulate them benignly in term to delay OA..On the basis of previous work, we plan to collect the blood of patients from the existing cohort, and prospectively follow up the patients for two years in order to explore the causal relationship between wind-cold-dampness affecting Thermo-TRPs/HSPs pathways and knee joint degeneration. We will use the animal model of wind-cold-dampness Bi syndrome to observe wind-cold-dampness affecting the state of Thermo-TRPs/HSPs and the targets of Guizhi Decoction. In the end, we will use animal serum of wind-cold-dampness to intervene chondrocyte, and explore the characteristics of wind-cold-dampness affecting Thermo-TRPs/HSPs crosstalk and the intervention mechanism of Guizhi Decoction. The research will help to reveal the material basis of the classical theory of traditional Chinese medicine and the scientific value of traditional Chinese medicine in treating OA.
