为应对细菌耐药性问题以及减缓抗生素研发压力,研发新型抗菌肽类药物成为人们关注的焦点之一。本课题组的前期研究发现,来源于黄边胡蜂的多肽Mastoparan-C (MP-C) 具有突出的抗菌活性,其结构的改变对其稳定性、抗菌效力以及毒性等生物学活性都有不同程度的影响。本项目选择MP-C为研究对象,采用氨基酸替换的方法进行长效、高效、低毒衍生物的构建,并鉴定这些衍生物的二级结构;通过一系列的生物学活性评价,分析衍生物的结构变化对其活性的影响;应用激光共聚焦实时成像技术结合β-半乳糖苷酶试验和多种荧光探针来探究不同的MP-C衍生物与模型菌的作用模式;应用转录组技术预测MP-C类多肽对细菌蛋白表达的影响。全面分析上述实验结果,阐明MP-C类多肽的构效关系,将为未来以MP-C为基础的新型抗菌兽药的研发奠定基础。
英文摘要
The development of antimicrobial peptides gradually become a research hotspot. The study on those peptides aims to reduce the stress of new antibiotics discovery which caused by the bacterial resistance. In our previous study, the structural-modifications of European Hornet-derived peptide Mastoparan-C (MP-C), which showed an obvious bacterial inhibitory function, will influence their stabilities, antimicrobial potencies and toxicities. Therefore, MP-C was selected as the research object in this project. Through the substitutions of amino acid residues, 4 peptide analogues were designed, followed by secondary-structural identifications. After that, the biological activities of those analogues will be compared to the natural, parent peptide for analysing changes in biological activities that were caused by structural alterations. Besides, the interaction modes of peptides on test strains could be explored through confocal real-time laser imaging technology and the ONPG test as well as several fluorescent probes. Meanwhile, the transcriptome analyses will be performed to predict the variances of peptide-induced expressions of bacterial intracellular proteins. Altogether, from former analyses, the structural-functional relationship of MP-C-like-peptide will be clarified. This study will provide theoretical bases for the constructions and modifications of further late-model veterinary antibacterial MP-C-like drugs.
