春化响应的分子机制是植物发育生物学的研究热点,春化过程可以激活冬小麦关键基因TaVRN1的表达而诱导开花,而TaVRN1转录激活的分子机制却知之甚少。我们前期的研究工作表明蛋白的O-GlcNAc修饰参与冬小麦春化响应关键基因TaVRN1基因的转录调控。O-GlcNAC糖基转移酶是否直接参与植物的春化响应,目前并不清楚。本项目拟通过分离冬小麦O-GlcNAc糖基转移酶TaSEC在春化响应过程中蛋白复合物的组分,研究TaSEC调控小麦春化响应关键基因表达的分子机制;同时拟分析小麦莽草酸激酶TaSK的翻译后O-GlcNAc修饰介导氨基酸代谢调控冬小麦春化过程的分析和细胞机制。该研究将有助于阐明O-GlcNAC信号调控春化关键基因TaVRN1表达参与春化响应的新的生物学功能,并为农作物花期调控的分子设计育种提供重要的理论指导。

The molecular mechanisms of vernalization response are hot topics in plant developmental biology, vernalization activates the transcription of the key regulator TaVRN1. But little is known about the pathways by which the key positive factor TaVRN1 is activated during vernalization process. We previously reported that post-translational O-GlcNAc modification of proteins is involved in TaVRN1 expression regulation; and the O-GlcNAc transferase SEC acts as a negative regulator of flowering to activate histone methyltransferase ATX1 by catalyzing its O-GlcNAc modification. It is not clear whether SEC is directly involved in vernalization response, and functions in a protein complex to regulate TaVRN1 transcription at a certain stage during vernalization process. In this project, we will identify the components in TaSEC protein complex to further analyze the molecular mechanisms of TaSEC regulating the expression of TaVRN1 in vernalization response. At the same time, we will study the molecular and cellular mechanisms of shikimate kinase TaSK involved vernalization regulation by O-GlcNAc signaling-mediated amino acid metabolism. The research findings of this project will elaborate the novel biological function of O-GlcNAc signaling in regulating vernalization response, and provide significant theoretical information for molecular design breeding of flowering time regulation in crops.
