黑肩绿盲蝽是稻飞虱的重要捕食性天敌,对田间稻飞虱数量的控制具有重要意义。提高黑肩绿盲蝽的繁殖力,将有助于提高其控害作用,但黑肩绿盲蝽生殖的激素调控机制研究甚少。前期研究表明蜕皮激素(20E)参与了黑肩绿盲蝽的生殖调控。本项目首先克隆20E信号通路关键转录因子Br-C的不同亚型,并分析各亚型的时空表达谱;采用RNA干涉技术、免疫组织化学等方法,研究Br-C各亚型在调控黑肩绿盲蝽卵黄发生、卵子生成和生殖力等方面的功能,筛选出对生殖具抑制或促进作用的亚型;再采用基因定点突变、细胞转染和EMSA等技术研究Br-C活性亚型对ClVg的转录调控,从而阐明Br-C不同亚型对黑肩绿盲蝽生殖的调控机制。本研究对阐明20E调控黑肩绿盲蝽生殖的作用机制,充分利用该天敌昆虫进行稻飞虱的生物防控、减少化学农药使用、实现稻田害虫的可持续治理具有重要意义。
英文摘要
Cytorhinus lividipennis is an important predator of rice planthopper, and plays a crucial role in controlling rice planthoppers in the paddy. Improving the fecundity of C. lividipennis will be helpful to improve its control effect on pests. However, little is known about the hormonal regulation mechanism of reproduction in C. lividipennis. Our previous study indicated that 20-hydroxyecdysone (20E) is involved in the reproductive regulation of C. lividipennis. In this project, we would firstly clone the informs of the key transcription factor Br-C (Broad-complex) of 20E signaling pathway in C. lividipennis, and analyze the spatiotemporal expression profiles of each inform. Subsequently, using RNA interfere and immunohistochemical technologies, we would examine the roles of each Br-C inform on the vitellogenesis, oogenesis, and fecundity of C. lividipennis. The informs that inhibit or promote reproduction would be screened. Finally, using site-directed mutagenesis, cell transfection and EMSA (Electrophoretic Mobility Shift Assay) technologies, we would examine the role of active Br-C informs in regulating the transcription of ClVg. Thus, the regulation mechanism of Br-C informs on the reproduction of C. lividipennis would be elucidated. This study will play great role on understanding the mechanism of 20E regulating the reproduction in C. lividipennis, making full use of C. lividipennis, and achieving an effective, sustainable pest management strategy for rice.
