Diet-induced excessive fat accumulation is the risk factor for metabolic imbalance. Recent studies have found that dietary polyphenols can induce white fat browning and promote fat consumption, which has become an effective strategy to improve metabolic imbalance, but its molecular mechanism has not been fully elucidated. The applicant recently obtained important clues that pomegranate polyphenols can induce white fat browning, and studied that the mitochondrial outer membrane protein mitoNEET may be involved in initiating this process, and the expression of mitoNEET may be regulated by miRNAs such as miR-155-5p. Therefore, this study hypothesized that pomegranate polyphenols can specifically regulate the level of miRNAs in white fat, thereby activating mitoNEET-mediated white fat browning. This study will first analyze the time-effect relationship of pomegranate polyphenols in regulating mitoNEET expression during the induction of white fat browning; Second, silence mitoNEET gene expression and analyze its role in pomegranate polyphenol-induced white fat browning; Finally, the screened miRNA was used to explore the molecular mechanism of pomegranate polyphenols regulate mitoNEET expression through miRNA. This study is expected to reveal the mechanism of pomegranate polyphenols in inducing white fat browning and provide a new theoretical basis for dietary polyphenols to improve metabolic imbalances.
