非酒精性脂肪肝病(NAFLD)系诱发肝癌风险的首要因素。防治NAFLD已是医学界对降低肝癌发病率的重要策略。肠道黏膜通透性增高是NAFLD发病主因。前期研究证实,揉腹法可重塑细胞骨架,组装锚定顶端连接复合体,从而降低肠道黏膜通透性。但决定通透性的另一关键要素肠上皮细胞(IEC)的调控机制尚待明确。本课题系前期项目的延伸,以揉腹法重塑细胞骨架为切入点,以诱导自噬发生,调控IEC凋亡进程为突破口,综合运用分子生物学、细胞生物学、病理学等方法研究揉腹法治疗NAFLD的作用机制。以期证实“揉腹法机械力重塑细胞骨架,特异性抑制Caspase-3/ROCK1/MLC信号通路级联反应,激活自噬启动激酶ULK1,诱导自噬体的形成及转输,从而调控IEC凋亡进程,降低肠道黏膜通透性,逆转肝细胞脂肪变性”假说的科学性,进一步揭示揉腹法防治NAFLD作用机制,为构建推拿防治消化系统疾病提供科学依据。

Non-alcoholic fatty liver disease (NAFLD) is the primary risk factor for liver cancer.Prevention and treatment of NAFLD has become an important strategy to reduce the incidence of hepatocellular carcinoma.The main cause of NAFLD is increased intestinal mucosal permeability.Previous studies have confirmed that abdominal rubbing can reshape the cytoskeleton, assemble and anchor the apical junction complex, thereby reducing intestinal mucosal permeability.However, the another key factor that the homeostasis regulation mechanism of intestinal epithelial cells (IEC)to maintain intestinal mucosal permeability remains to be clarified This topic is an extension of the previous project of the department.This project aims to induce autophagy and regulate the apoptotic process of IEC,and taking the abdominal rubbing method to remold the cytoskeleton as the breakthrough point.Molecular biology, cell biology and pathology were used to study the mechanism of abdominal rubbing in the treatment of NAFLD.In order to confirm that "abdominal rubbing mechanical force remodels the cytoskeleton, specifically inhibits caspase-3/ROCK1/MLC signaling pathway cascade reaction, activates autophagy-activated kinase ULK1, induces autophagy formation and transfusion, thus regulating the process of IEC apoptosis, reducing intestinal mucosal permeability and reversing hepatocyte steatosis"hypothesis scientificality. To further reveal the mechanism of abdominal rubbing in the prevention and treatment of NAFLD, and to provide scientific basis for the construction of massage in the prevention and treatment of digestive system diseases.
