CRISPR/Cas9介导获得双峰驼诱导多能性干细胞

本项目拟在课题组已具备的基因克隆与分析平台、干细胞分离建系平台、CRISPR/Cas9基因编辑平台、各种组学生物信息学分析平台以及细胞核移植（克隆）平台基础上，围绕着利用CRISPR/Cas9系统生产双峰驼诱导性多能干细胞（iPSc）的问题，重点解决稳定传代双峰驼iPS细胞系中外源基因整合位点筛选；双峰驼iPS细胞特异表达基因的筛选；新诱导基因的筛选；比较双峰驼iPS细胞系与通过CRISPR/Cas9系统获得的iPS细胞系之间生物学特性的异同，以期获得双峰驼生殖生理的相关基础资料，丰富动物胚胎干细胞和发育生物学的内容。这对保护动物的遗传资质、提高iPS细胞诱导效率以及发展西北地区生态农业具有重要的实际意义。

This project is based on the platform of gene cloning and analysis , stem cell isolation ,CRISPR/Cas9 gene editing ,various bioinformatics analysis and nuclear transfer (cloning) ,surrounding the problem of obtained induced pluripotent stem cells (iPSc) from Bactrian camels via the CRISPR/Cas9 system , focusing on the screening of exogenous gene integration sites in stable passaged Bactrian camel iPS cells line; screening of iPS cells-specific expression genes in Bactrian camel Screening of newly induced genes; comparing the similarities and differences of biological characteristics between the iPS cell line of Bactrian camel and the iPS cell line obtained by CRISPR/Cas9 system, in order to obtain the basic data of reproductive physiology of Bactrian camel, enrich animal embryonic stem cells and developmental biology .This has important practical significance for protecting animal genetic qualification, improving iPS cell induction efficiency and developing ecological agriculture in Northwest China.
