微波辐射对人体健康尤其是儿童大脑发育的损害日益引人关注,而微波辐射致海马神经发生障碍及其在认知损伤中的作用研究尚属空白。本项目聚焦的关键科学问题是:miR-17-92 cluster介导微波辐射损伤幼年海马神经发生的病理机制及干预策略。我们前期研究发现,30mW/cm2微波辐射可损伤与海马神经发生微环境密切相关的齿状回亚颗粒区,抑制突起生长和树突棘成熟,下调miR-17-92 cluster的表达。鉴此,本项目拟利用RNA-seq筛选miR-17-92 cluster的上游调控基因及靶基因,研究miR-17-92 cluster及其靶基因PTEN及Tbr2通过调节神经前体细胞增殖及分化的关键分子SOX2、相关信号通路(PI3K/AKT/mTOR、Shh、Wnt)参与微波辐射致海马神经发生障碍的分子机制。以期揭示微波辐射致认知功能障碍的分子机制,为探寻敏感指标和防治靶点奠定基础。
英文摘要
The widespread use of microwaves has raised questions about its adverse effects on human health, especially on juvenile brain development. However, the mechanism of hippocampal neurogenesis disorders induced by microwave and its role in cognitive impairment have not yet been elucidated. The critical scientific issue focused on this project is the miR-17-92 cluster mediates the pathogenesis of juvenile hippocampal neurogenesis impairment induced by microwave exposure and the interventional strategy. Our preliminary study indicated that 30mW/cm2 microwave exposure could damage the dentate gyrus sub-granular zone (hippocampal neurogenesis niches), inhibit the growth of dendrites and the maturation of dendritic spines, suppress the expression of miR-17-92 cluster in hippocampus. Therefore, this project intends to identify upstream regulatory genes and target genes of miR-17-92 cluster by RNA-seq in hippocampus after microwave exposure. And to further reveal the mechanisms of miR-17-92 cluster and their target genes (PTEN, Tbr2) involved in hippocampal neurogenesis disorders induced by microwave by regulating key molecules SOX2 and related signaling pathways (PI3K/AKT/mTOR, Shh, Wnt) of neural progenitor cell proliferation and differentiation. Therefore, our project will contribute significantly to both understanding molecular mechanisms of cognitive dysfunction induced by microwave and finding novel signatures or effective therapy for microwave-induced brain injury.
