植物雄性不育性已进行了较多研究,克隆了许多不育相关基因。但这些基因是如何通过mRNA表达的蛋白质发挥作用的还不是很清楚。因此,联合比较分析不育株和可育株间的差异表达mRNA和蛋白质,对深入了解雄性不育机理具有重要的作用。本研究以自育的晒烟胞质雄性不育系及其保持系为材料,在烟草花蕾发育的关键时期,提取两系花蕾的总RNA和蛋白质,构建RNA测序文库并测序,分析测序结果获得差异表达mRNA(基因);花蕾蛋白质经胰蛋白酶消化后进行iTRAQ标记及质谱检测分析,获得差异表达蛋白质。对差异表达mRNA和蛋白质进行相关分析,获得两者有相同表达趋势的基因,从中选择与烟草育性密切相关的基因构建表达载体转化烟草,验证其与雄性不育性的关系,以期深入了解烟草雄性不育的分子机理。这将为揭示烟草雄性不育的分子遗传机理、深化烟草雄性不育杂种优势利用的理论和应用研究奠定基础;同时也可对其他作物雄性不育机理的研究提供参考。

More and more researches had made in the plant male sterility, many male sterility related genes or gene segments have been cloned. But how do these genes play its role effectively by means of the expression of mRNA and proteins is not very clear. Correlation conjoint analysis of the differential mRNA and proteins in the critical period of bud development between the male sterility and fertile plants plays an important role in further understanding the mechanism of male sterility. With the purpose of giving an insight into the molecular mechanism of tobacco male sterility, the self-bred sun-cured tobacco(N.Tabacum) cytoplasm male sterility line and its maintainer line will be taken as test materials in this research. In the key period of tobacco bud development, sterility and fertility buds would be taken to extract mRNA and proteins. RNA sequencing library will be constructed and sequenced, and the sequencing results would be analyzed to obtain differentially expressed mRNA(gene). The extracted proteins will be labeled with iTRAQ after digested by trypsin, the labeled proteins will be detected by mass spectrometer after graded by strong ion chromatographic column. The protein expression quantity will be obtained and analyzed according to the result of mass spectrometry identification. Genes with the same expression trend of the differentially expressed mRNA and protein would be obtained by correlation analysis of differentially expressed mRNAs and proteins. In order to verify the relationship between these genes and tobacco sterility and understand the molecular mechanism of male sterility in tobacco, the plant expression vector of the representative different genes would be constructed and be to transform the tobacco. All these works will lay a foundation for revealing the molecular genetic mechanism of tobacco male sterility and for deepening the theory and application research of tobacco heterosis utilization with CMS, of which also provide a reference to the mechanism of other plant male sterility.
