抽穗期是一个决定小麦区域适应性和分布的重要农艺性状,受多基因调控,春化基因是调控抽穗期主要遗传因子之一。CASL3AL是一个中国春(CS)-野生二粒小麦(TTD)染色体臂置换系,其3AL染色体臂被TTD置换。前期的研究发现,CASL3AL在多个环境都比CS早熟且对春化更敏感,并在3AL的barc324与P1381标记间定位到了一个主效QTL,表明在CASL3AL的3AL上有一个新的对春化敏感的基因位点,命名为QHd-3AL。在此基础上,本研究将利用含有目标QTL的小片段材料与CS杂交构建F2大群体及其F2:3次级分离群体对QHd-3AL进行精细定位;通过CASL3AL重测序和转录组测序分析,结合生物信息学分析方法筛选关键候选基因,利用qRT-PCR和VIGS技术对其进行初步验证。本研究结果将丰富小麦春化基因调控开花的理论体系,同时为选育小麦早熟高产新品种提供理论参考和材料基础。
英文摘要
Heading date, regulated by polygenes, is one of the important agronomic traits determining the regional adaptability and distribution of wheat. Vernalization gene is one of the main factors regulating heading date. CASL3AL is a chromosome-arm substitution line (CASL) of wild emmer (TTD) in the background of common wheat variety Chinses Spring (CS), in which the long arm of chromosome 3A of CS is replaced by the corresponding chromosome of TTD. Previous studies of our laboratory showed that CASL3AL headed earlier and was more sensitive to vernalization than CS in different environments, and a significant QTL was detected in the region of barc324-P1381 on 3AL, indicating a new vernalization-sensitive gene locus on 3AL of CASL3AL, named QHd-3AL. Based on this basis, the aims of this study are to fine map the vernalization gene on 3AL in CSAL3AL using a large F2 population and its F2: 3 secondary segregated population derived from the cross between the small fragment material containing target QTL and CS, combining with the constructed RILs population, to screen the key candidate gene through the analysis of CASL3AL resequencing and transcriptome sequencing, integrating bioinformatics analysis methods, and to preliminarily verified the candidate gene using qRT-PCR and VIGS technology. The results of this study will enrich the theoretical system of wheat vernalization genes regulating flowering, and provide theoretical reference and material basis for breeding new wheat varieties with early maturity and high yield.
