由于滥用和非法使用违禁药物,造成动物源性食品磺胺类兽药残留超标问题比较突出。标准的磺胺类药物残留检测如液相色谱-串联质谱法虽然方法灵敏、准确,但仪器贵重、检测成本高、操作繁琐等不适于现场检测,因此,发展低成本、方便携带、可靠的现场检测方法是监控兽药残留的重要手段之一。本研究利用铂(Pt)微电极作为基底电极,简化三电极为两电极系统,针对其电催化活性低和选择性差的不足,利用多壁碳纳米管(MWCNTs)高比表面积、高电导率特性和Nafion对阳离子的选择性,制备嵌入式Nafion-MWCNTs粉末微电极(PME),既能克服常规电极受到稳态的制约,又可实现在线清洗,避免长期使用带来的稳定性低、重复性差等。利用制备的嵌入式Nafion-MWCNTsPME,基于暂态电化学响应机理,结合自行设计的便携式电化学检测装置,建立一种适用于现场快速检测磺胺类兽药残留的方法。

Due to abuse and illegal use of drugs, the super standard of sulfonamides residues in animal derived food was more outstanding. The standard detection method for sulfonamides residues such as liquid chromatography-tandem mass spectrometry was sensitive and accurate, but this technique required large valuable instrument and the detecting procedures were high cost and complicated etc., was not suitable for the field detection. Therefore, the development of field detection method as well as advantages of low cost, easy to carry and reliable was very important to monitor food veterinary drug residues. In this study, platinum (Pt) microelectrode was used as basal electrode, the three electrodes systems could be simplified as two electrodes systems. In view of its low electricity catalytic activity and poor selectivity, based on the high specific surface area, high conductivity of multi-wall carbon nanotubes (MWCNTs) and Nafion cationic selectivity, the embedded Nafion-MWCNTs powder microelectrode (PME) was fabricated. This PME could overcome the restriction of the steady-state for inconventional electrodes, and could realize online cleaning, avoid long-term use of low repetitive and poor stability etc. According to transient electrochemical principles,using the embedded Nafion-MWCNTsPME, which combined with the home-made portable electrochemical detection device, a new method for field detecting sulfonamides residues was established.
本项目针对动物源性食品磺胺类兽药残留超标问题比较突出,而目前标准的磺胺类药物残留检测仪器贵重、操作繁琐不适于现场检测,利用电化学检测易于易与其它测试、控制技术联用、仪器容易小型化的优势,从微观水平设计电极功能,将不同功能化处理的纳米材料引入电极表面,构建基于纳米结构电极的快速检测磺胺类药物的方法。具体成果包括:首先,采用混酸液相氧化法和超声分散法制备出羧基化DMF-MWCNTs,利用物理吸附法制备出羧基化DMF-MWCNTs/GCE和羧基化DMF-MWCNTs/ME修饰电极,成功用于磺胺类药物的快速检测研究。其次,采用表面沉积交联法制备出功能化纳米复合材料Nafion-MWCNTs,利用化学腐蚀法和研压填充法制备了嵌入式Nafion-MWCNTs/PME电极,简化三电极为两电极系统,实现了对磺胺类药物残留的快速检测,克服了常规电极受到稳态的制约,避免了电极长期使用带来的稳定性低、重复性差等问题。本项目的研究为食品安全检测现场化、小型化的实现提供有力支持。同时也构建了其它纳米电化学传感器用于维生素C、呋喃西林及其代谢物等的检测研究,拓宽了功能化纳米材料的应用领域。
