The intrauterine infection of human cytomegalovirus is closely related with abortion, fetal death, teratism, premature delivery and mental retardation, visual retardation, dysaudia ect. Ganciclovir has an ideal effect of anti- HCMV, but whose usage is restrict in the gestation period。Since the guinea pig's gestation period and placenta construction are similar to human's, the intrauterine infection of guinea pig cytomeglovirus (GPCMV) also resemble the intrauterine infection of human cytomeglovirus(HCMV). Abroad scholars had established the animal model of the intrauterine infection of GPCMV, which had used to research the pathogenesis of HCMV intrauterine infection and screening vaccines, but had not applied it to the research of anti-virus medicine. 2) Research direction: The members of this item have proved by in vitro experiment that the Chinese herbal medicine Jinyrbaidu has the function of anti- HCMV. On the level of the whole animal, this item has researched the function of anti-GPCMV and it would establish the theory background for the clinical research..3) Main contents: ① Establish the experimental animal model of the intrauterine infection of GPCMV.② Establish the effects of Chinese herbal medicine Jinyebaidu in prevention and cure the intrauterine infection of GPCMV and its main valid composition by animal model. ③The reliability of the medicine used in gestation period was established through the breeding toxicity experiment. 4) Important results and key data: ①Among the 30 gestation guinea pigs in the blank control group, the abnormality(such as conceptus loss, fetal resorptions etc.) rate of offspring is 6.71%(9/134), however, among the infection group (30 gestation guinea pig) which had inoculated 107 TCID50 GPCMV intraperitoneal, the abnormality rate of offspring is 61.22%(60/98), there is significant tatistic difference.(x2=80.480 P=0.001). ②The positive rate of GPCMV DNA in the maternal blood of the infection group is 100%(30/30), and the offspring infection rate is 100%(98/98).Among the guinea pig which had been inoculated GPCMV and offered Jinyebaidu subsequently, the infection rate of parental generation and offspring are 53.33%(16/30) and 51.24%(62/121) respectively, and the abnormality rate of offspring is 28.10%(34/121), Between the two groups, there is very significant difference. (x2=18.261 P=0.001, x2=65.406 P=0.001, x2=80.480 P=0.001).Comparing the influences of four main compositions of Jiyebaidu for the infection rates of parental generation and offspring, the differences have not show the significance. (P>0.05).Given Jiyebaidu in the organogenetic period of mice, we did not discovers it showed significant bad influence to reproductive system of parental generation and the growth of offspring and it had the function of adjust the cellular immunity function of pregnant mice. 5) Science meaning: It is the first time that the experimental animal model of the intrauterine infection of GPCMV was established at home, and it is also the first time that the experiment animal model was utilized to proceed the anti-CMV research of Chinese herbal medicine and the infection of offspring at home and abroad.The first time, it had been proved by the animal experiment that Chinese herbal medicine jinyebaidu had the important clinical application prospect since it had the function of decrease the infection rates of parental generation and offspring and alleviating infection symptoms of offspring and safety in the gestation period.
建立豚鼠巨细胞病毒胎仔感染动物模型,用分子生物学、病毒分离及病理学等方法检测胎仔血液、内脏、胎盘巨细胞病毒受累状况及病理变化,观察感染对胎仔生长发育的影响;在动物模型上研究中医药金叶败毒颗粒防治胎仔感染的效果及其主要有效成份;并进行其生殖试验。为临床应用中医药进行孕期治疗、防治宫内感染,提供理论依据。
