抗氧化活性是衡量果蔬品质的重要指标,如何维持和增强果蔬采后抗氧化活性是果蔬贮运领域亟待解决的技术瓶颈难题。糖信号转导途径是当前研究糖调控果蔬采后保鲜的新兴热点问题。我们前期研究表明,荧光照射诱导采后蔬菜抗氧化活性和糖累积,糖累积对抗氧化活性有显著调控作用,但机制不明。本项目以离体叶片和叶圆片为试材,采用糖类似物法,从信号转导途径揭示糖累积对采后叶片抗氧化活性的调控机制。重点从依赖和不依赖己糖激酶(hexokinase,HXK)信号转导途径比较糖和糖类似物是否通过己糖激酶途径行使糖信号功能,激发下游糖信号参数(抗氧化物质、酶、基因等)应答,探讨HXK抑制剂能否阻断糖信号途径并相应地解除糖对上述参数的调控作用,从而在物质代谢、酶活性、基因表达水平上阐明光照调控采后叶片抗氧化活性的糖信号依赖途径和分子机制。研究结果将揭示光照增强采后果蔬品质新机制,也为研究可溶性糖调控果蔬采后保鲜提供新思路。
英文摘要
Antioxidant activity (AA) is an important index on estimating fruits and vegetables quality. How to maintain and improve AA of postharvest fruits and vegetables is a critical a technical bottleneck in postharvest fruits and vegetables field. In addition, sugar signal transduction pathway is a hot scientific issue of sugar regulating postharvest fruits and vegetables preservation. Our previous studies showed that fluorescence irradiation induced AA and sugar accumulation in postharvest vegetables. Sugar accumulation was positively related to AA, but the mechanism remained unclear. The aim of this project is to reveal the mechanism of sugar accumulation regulating AA of postharvest green vegetables through signal transduction pathway. The project mainly study that sugar and sugar analogs trigger the responses of sugar signaling parameters (antioxidant substances, enzymes, and related genes) by dependent and/or independent hexokinase (HXK) signaling transduction pathway. In addition, the study also investigates whether HXK inhibitors block the sugar signaling pathway and release the regulation of sugar on these parameters accordingly. Thus the sugar signal transduction pathways and molecular mechanisms of fluorescent light irradiation regulating AA of postharvest leaves will be elucidated on substance metabolism, enzyme activity, and genes expression level. The result obtained is expected to develop the novel theory of fluorescent light irradiation regulating the postharvest quality of fresh fruits and vegetables. At the same time, this research will offer a novel research thinking about sugar preserving postharvest quality of fruits and vegetables.
