以抗体为核心试剂的免疫分析技术是环境与食品安全监测的重要手段;但抗体仍存在特异性低、稳定性差等问题,不能满足应对突发事件抗体快速制备的需求;而VH:VL来源于同一B细胞的天然成对抗体在稳定性和特异性上具有先天优势。本项目拟以典型环境与食品有害物多氯联苯、苯并芘与四环素等多种危害物为靶标,利用乳液PCR(emulsion PCR)高效获取天然成对抗体基因,建立酵母展示天然成对抗体库,系统研究天然成对抗体库的设计与构建、筛选优化等,多轮高通量筛选天然成对抗体并在哺乳动物细胞中进行高效表达,以表面等离子体共振等技术评价和鉴定抗体效价、结合活性和特异性、稳定性等,最终实现高稳定性、高特异抗体的快速筛选与制备。研发环境与食品安全危害物特异抗体的快速筛选技术,不仅是抗体制备新技术的有益探索,还可为环境与食品安全事件迅速提供关键试剂,在进出口检验检疫、卫生监督检测、企业质量控制等领域具有广阔的应用前景。

Antibody-based immunoassay is an important method of monitoring environment and food safety. However, there are various problems during the preparation of antibody, such as the low specificity and poor stability of antibody, which cannot meet the needs of emergency detection antibodies. The VH:VL natively paired antibody have the innate advantages of specificity and stability because of the genes of VH and VL derive from same B lymphocytes. By using emulsion PCR to realize the rapid acquisition of natively paired antibody genes and using yeast display technology for effective activity display and rapid screening, this project would develop yeast display paired antibody library and systematically study the design and construction of yeast display natively paired antibody library and the optimization of screening conditions. Targeted the typical environmental and food contaminants such as PCBs, benzopyrene and tetracycline, the natively paired antibodies would be selected after high-throughput screening and be highly expressed in mammalian cells as monoclonal antibodies. The characters of the antibody, antibody titer, binding activity, specificity and stability, will be evaluated and identified by surface plasmon resonance (SPR), ELISA and other methods. Finally, this project will realize the rapid screening and preparation of highly sensitive and stable antibody. The research and development of rapid screening technology for specific antibodies is not only a beneficial exploration of the preparation of antibodies against hazards, but also can provide a key reagent for food safety detection. It has broad application prospects in the fields of import and export, food hygiene supervision and inspection, quality control and so on.
