T细胞分化引起的不同细胞免疫应答,在获得性抗病毒免疫中发挥着关键作用,其中T细胞的定向分化和效应是抗原特异的MHC和TCR分子以及细胞微环境相互协同作用的结果,但由于这些分子基因的遗传多样性和互作因子的复杂性,基于病毒抗原免疫特异的猪MHC介导的T细胞定向分化的机制研究在国际上尚属空白。项目以猪瘟C株疫苗病毒抗原免疫不同遗传背景的近交系小型猪,分选和鉴定免疫特异的DC、CD4/CD8T细胞不同亚群,通过基因扫描和单细胞测序分析与克隆优势SLAⅠ、SLAⅡ类分子和αβTCR基因,建立APC与T细胞互作模型研究机体T细胞定向分化和应答特性与规律以及驱动因素,解析猪瘟病毒抗原免疫特异的SLA介导的αβT细胞定向分化的内在本质,揭示其遗传多样性与免疫学功能特异性的分子机制,为猪细胞免疫机制及高效疫苗产品研发提供理论依据。
英文摘要
Different cellular immune response caused by T cell differentiation, play an important role in host acquired cellular immunity to against viral infection, of these results that directional differentiation and reaction of T cells is co-controlled by antigen-specific MHC,TCR and cytokines bewteen cell. but investigation of T cell directional differentiation driving by swine MHC(SLA) in pig immunized by viral antigen is still not report due to the diversity and complexity of SLA and TCR. This firstly sorting and identify the DC and CD4+/CD8+ T cell subgroup with flow cytometry, and amplify, clone and analysis the repertoires of SLAⅠ, SLAⅡ and αβ TCR gene with Genemapper scaning and single cell sequencing by immunized antigen of C strain-swine fever virus in different inbred minipigs. Then establish a model of interaction between APC and T cell, and analyze the genetic constitution, structural feature and its diversity and specifity of SLAⅠ, SLAⅡ and αβTCR, and highlight the rule of cloning proliferation, directional differentiation and driving intrinsic factors of predominated usage of SLA and TCR. Thirdly, reveal the molecular mechanism of genetic diversity and functional specifity which provided theoretical basis and technical support for development of vaccine and mechanism of cellular immune response in swine.
