雄性昆虫精子的形成在昆虫种群繁衍过程中具有不可或缺的作用,寻找调控精子发生的关键基因特别是突变后导致雄性不育的基因成为昆虫不育技术研究的焦点。我们在家蚕种质资源中新发现一个雄性不育的突变品系,该品系的雌性是正常可育的,而雄性成虫虽然能够与雌性成虫正常交配,但交配后的雌性产不受精卵,经典遗传分析表明该突变体由一个隐性基因控制,命名为gms。家蚕是鳞翅目模式昆虫,该突变体为研究确定家蚕精子发育关键基因提供了良好的模型,本项目以该雄性不育突变体为研究对象,通过图位克隆的方法寻找突变体形成的关键基因,利用CRISPR/Cas9技术验证该关键基因的功能;采用转录组测序技术、免疫组化等分子生物学技术分析雄性精子形成的信号通路,初步阐明该突变体雄性不育发生的分子机制,为家蚕品种选育和繁殖提供理论指导,为应用昆虫不育技术生物防治鳞翅目农林害虫提供合适的靶基因。
英文摘要
The formation of sperm of male insects is of great importance in insect population reproduction. The hotspot of insect sterility research is focused on seeking for key genes that regulating sperm formation, especially those genes that mutations result in male sterility. We have found a novel mutant of male sterility from silkworm Bombyx mori resource preserved. Females of this mutant are normal and fertile. While its males are abnormal although they can mate with females, but females lay unfertilized eggs after mating with males of the mutant. Classical genetic analysis showed this mutant is controlled a recessive gene named gms. B. mori is a model insect of Lepidoptera, and this mutant provides an excellent model for study of key genes of sperm development. In this project, we are going to use this male sterility mutant as the subject, to search for key genes related to formation of the mutant by positional cloning; to study the function of the key gene by CRISPR/Cas 9 technique; to analyse the signal pathway of male sperm formation by RNA-seq, immunohistochemistry techniques etc. Thus to clarify the molecular mechanism of male sterility of the mutant, and provides theoretical guidance for breeding of B. mori varieties, as well as a suitable target gene for utilization of insect sterility technique in control of Lepidoptera pests.
