作为一种新兴有机污染物,四溴双酚A(TBBPA)已在渤海无脊椎动物体内被广泛检出,但对其生殖内分泌干扰的相关研究几乎空白。申请人前期研究发现,TBBPA暴露可导致紫贻贝配子发生及信号转导等过程异常,推测存在生殖内分泌干扰效应。为进一步探究TBBPA对海洋双壳贝类的生殖内分泌干扰效应及其作用机制,本项目拟利用贝类生理学研究手段分析TBBPA对紫贻贝性腺发育、配子发生以及受精等生殖行为的影响,揭示TBBPA的生殖毒性;同时,借助转录组学和生化指标分析,解析TBBPA对紫贻贝生殖内分泌的干扰途径,聚焦类固醇性激素合成、代谢、非基因组效应以及生殖、发育等过程,筛选生物标志物;通过对生物标志物与异常生殖行为指标进行相关性分析,阐明TBBPA对紫贻贝生殖内分泌干扰效应的作用机制,填补TBBPA对海洋无脊椎动物生殖内分泌干扰研究的空白,为评估渤海TBBPA污染的生态风险提供科学依据。
英文摘要
As an emerging organic pollutant, Tetrabromobisphenol A (TBBPA) has been found to be accumulated in marine invertebrates from the Bohai Sea. However, few studies on the reproductive endocrine disruption by TBBPA have been conducted on marine invertebrates. Previous investigations by our research group revealed that gametogenesis and signal transduction in mussel Mytilus galloprovincialis were significantly influenced by TBBPA, implying the endocrine disrupting effect of TBBPA. In order to verify the hypothesis, this project will uncover the reproductive toxicity of TBBPA by evaluating the impacts on reproductive activities, including gonad development, gametogenesis, and fertilization. Meanwhile, we will use transcriptomics as well as biochemical analyses to unravel the mode of action of endocrine disruption by TBBPA in mussel M. galloprovincialis. The responsive genes and enzymes that are closely related to sex steroids biosynthesis, metabolism, signaling pathway, reproductive, and development will be selected as potential biomarkers of TBBPA. Finally, the correlation analysis will be performed between biomarkers and abnormal reproduction, aiming to illuminate the mechanisms linked to endocrine disrupting effect of TBBPA. This project focusing on endocrine disrupting effect of TBBPA in marine invertebrate for the first time will hopefully provide scientific basis for ecological risk assessment of TBBPA pollution in the Bohai Sea.
