自20世纪80年代以来,人类对衰老表现出了强烈兴趣。利用经典的Horvath 表观遗传时钟计算DNA甲基化年龄(DNAm年龄),从分子水平来研究衰老的影响因素及其发生发展机制更是近年的研究热点。研究表明,吸烟饮酒、膳食、体力活动等行为生活方式是DNAm年龄的重要影响因素。而同卵双生子是研究表观遗传学问题的绝佳对象。基于此,本研究拟利用中国双生子登记系统筛选232对同卵双生子,通过问卷调查、甲基化检测、Horvath表观遗传时钟计算DNAm年龄,探索吸烟、饮酒、膳食、体力活动等行为生活方式与DNAm年龄及年龄加速的关联,以发现对DNAm年龄加速有较大影响的行为生活方式,从而进行干预,进而有效预防、减缓甚至逆转衰老。同时可为临床上衰老相关疾病的预测、筛检、诊断、预后等提供科学证据。这是国内首次利用同卵双生子进行DNAm年龄研究,从分子水平更深入地理解衰老机制,提供中国人群证据,追赶国际水平。

Since the 1980s, humans have shown a strong interest in aging.The classical Horvath epigenetic clock was used to calculate DNA methylation age (DNAm age) ,so as to study the influencing factors of aging and its mechanism of occurrence and development in molecular level.The research shows that smoking, drinking, diet and physical activity are the important influencing factors of DNAm age.Identical twins are an excellent target for the study of epigenetics.Based above, this study intends to screen 232 pairs of identical twins in the Chinese twin registry , by acquiring information in the questionnaire, methylation detection, DNAm age calculated by Horvath epigeneticclock DNAm age, to explore the association between DNAm age,DNAm age accelerated and behaviors such as smoking, drinking, diet and physical activity lifestyle and so on,to find the lifestyle that DNAm age accelerate has a great influence on ,to intervent the changeable lifestyle , so as to effectively prevent, slow down or even reverse aging.At the same time, it can also provide scientific evidence for clinical prediction, screening, diagnosis and prognosis of aging related diseases.This is the first time to use identical twins to study DNAm age in China, so as to better understand the aging mechanism at the molecular level and provide evidence for the Chinese population to catch up with the international level.
