CPD gene encodes C-3 oxidase, which plays a vital role in Brassinosteroids (BRs) biosynthesis. BRs play an important role in plant resistance to abiotic stress. However, the mechanism of CPD gene regulating the synthesis of BRs in response to salt stress in potato is still unclear. In the previous study, we found that the expression levels of CPD gene in potato were correlated with water stress. Therefore, CPD-overexpressing and interference-expressing (RNAi) transgenic potato plants will be used as experimental materials under salt stress, to excavate the upstream and downstream regulatory genes and proteins of the CPD gene, and differentially expressed genes and proteins in response to salt stress. The roles of the genes and proteins in the regulation of BRs response to salt stress will be explored. The effect of the expression level of CPD gene on the content of endogenous BRs will be also studied. And the key transcription factors regulating salt stress and CPD gene will be measured. Meanwhile, interaction between the key transcription factors and CPD gene in BRs synthetic process will be detected, and the interaction protein of CPD protein will be screened. The role of CPD in the response of potato BRs to salt sress will be analyzed, and the mechanism of CPD protein regulating the synthesis and accumulation of potato BRs will be revealed. The research will provide a theoretical basis for genetic improvement of potato salt tolerance.
