非酒精性脂肪性肝炎(NASH)的发生是肝脏固有免疫细胞所继发的炎症反应等一系列因素所导致,但缺乏针对性干预手段。肠道菌群变化通过影响胆汁酸成份和合成,介导CXCL16-CXCR6轴的激活,可能与NASH肝脏炎症反应的发生进展密切相关。我们前期研究发现益气补肾调脂方能够明显改善高脂高果糖饮食诱导NASH小鼠肝脏脂肪变和炎症程度。在此基础上,本研究拟通过建立小鼠NASH模型和体外脂肪肝模型,从分子、组织、基因等水平,首先明确益气补肾调脂方通过抑制肝脏固有免疫细胞浸润和活化减少肝脏炎症反应在NASH中的治疗作用,其次,阐明益气补肾调脂方通过调控胆汁酸/CXCL16/CXCR6轴抑制肝脏炎症反应的机制。通过本研究为进一步揭示益气补肾调脂方的具体作用靶点和调控环节,为明确益气补肾调脂方治疗NASH的疗效机制提供实验依据。

The occurrence of nonalcoholic steatohepatitis (NASH) is caused by a series of factors such as inflammatory response of liver innate immune cells, howerer,there is no targeted intervention.The intestinal flora changes affect the bile acid composition and synthesis, and mediate the activation of the CXCL16-CXCR6 axis, which may be closely related to the development of NASH liver inflammatory response.Our previous study found that Yiqi Bushen Decoction can obviously ameliorate hepatic steatosis and degree of inflammatory in NASH mice induced by high-fat and high-fructose diet.On this basis,at the level of molecule,tissue and gene, firstly, to clarified the therapeutic effect of Yiqi Bushen Decoction on hepatic inflammatory response of NASH by inhibiting the infiltration and activation of liver innate immune cells.Secondly, to elucidate the mechanism of Yiqi Bushen Decoction in inhibiting liver inflammatory response by regulating bile acid/CXCL16/CXCR6 axis.Our study is further to revealing the specific targets and regulatory links of Yiqi Bushen Decoction treatment of NASH,and providing the experimental basis for clarifying the therapeutic mechanism of Yiqi Bushen Decoction on NASH.
