慢传输型便秘(STC)是影响身心健康的常见病、多发病,结肠动力障碍是其重要病理特点。前期研究发现针刺可通过调节5-HT分泌改善结肠动力进而治疗STC。最新研究表明机械离子通道Piezo2是肠嗜铬细胞(EC)分泌5-HT的力化学耦合关键,针刺调节5-HT分泌改善结肠动力的效应是否通过EC力化学耦合值得深入探讨。本项目拟以针灸优势病种STC为研究对象,围绕“针刺可能通过调控EC细胞Piezo2—5-HT分泌的力-化学耦合路径,改善结肠动力进而治疗STC“这一假说,采用免疫蛋白组化、激光共聚焦荧光显微技术、模式动物(基因敲除)等多学科技术,以5-HT释放量、Piezo2结肠表达,5-HT与Piezo2共表达,结肠动力为主要观察指标进行正向评价,引入模式动物Piezo2CKO进行反向验证,综合探讨针刺改善STC结肠动力的作用机制,为针刺治疗动力障碍型胃肠疾病的临床运用提供更多科学依据。

As the most common disease, slow transmission constipation (STC) affects people's physical and mental health,and colonic motility disorders is an important pathological feature of STC. Previous studies have indicated acupuncture can be effective in improving colonic motility to treat STC by adjusting 5-HT secretion. Recent studies clearly indicated that Piezo2 ion channels are critical determinants of enterochromaffin cell (EC) in mechanochemical coupling. Among them, it is worthy of further exploration that the effect of acupuncture improving colonic motility to treat STC by adjusting 5-HT secretion is highly associated with mechanochemical coupling of EC.This project will take STC as the object which is an advantage of disease for acupuncture treatment, and we hypothesis that improvement the colonic motility of acupuncture is based on the mechanochemical coupling way of regulating Piezo2—5-HT secretion of EC. We use different technical approaches such as immunohistochemistry, laser scanning confocal fluorescence microscopy, knockout animals, to observe indicators of 5-HT release,expression of Piezo2 protein/gene, co-expression of 5-HT and Piezo2,evaluation of colonic motility for forward evaluating the hypothesis. And we use Piezo2CKO mice for reverse validating. Above all, this project is designed to investigate underlying mechanisms of acupuncture in the treatment of STC and to provide evidences for the clinical application of acupuncture in the treatment of gastrointestinal diseases.
