内生菌与药材品质密切相关。然而内生菌是潜在致病菌,植物维持内生菌无害时引起的药材质量变化尚不明确。在完成NSFC课题(No.81573537)中,从30株菌选出11株非致病菌,6株菌提高了产量或部分成分含量,但组合接种未见明显叠加效应,各组丹参酮含量均较商品药材低。我们推测内生致病菌有利丹参酮积累,遂将19株致病菌田间接种,6株菌提高了丹参酮含量。据此提出“环境驱动植物全生物跨界重组诱导防御代谢重编程以维持内生菌利害平衡从而影响药材质量”假说。本项目用4种缺素和2种气候胁迫组合模型,以根系选择培养,从内生真菌、细菌、放线菌和致病菌逐层组合,利用 Illumina、LC-MS和ICP-MS等技术跟踪各菌丰度、代谢和根际分泌物,厘清胁迫-全生物构成-生理代谢-根际效应-成分间关系。揭示响应环境驱动全生物关联药材质量变化机制,获得中药品质形成机制新见解,提供药用植物生态栽培研究新思路和和新方法。
英文摘要
The endophytic bacteria is closely related to the quality and geoherbalism of medicinal materials.However, endophytes are potential pathogens, and it is not clear that the quality of medicinal materials changes when plants maintain endophytes harmless.In the NSFC project (No.81573537), 11 non-pathogenic bacteria were selected from 30 strains, and 6 strains could improve the yield or active component content partly , but there was no obvious superposition effect in combination inoculation, and the content of tanshinone in each group was lower than that in commercial medicinal materials. It was speculated that endogenous pathogenic bacteria were involved in tanshinone accumulation, so 19 strains of pathogenic bacteria were inoculated in the field, and 6 strains could improve tanshinone content.Therefore, the hypothesis are proposed that transboundary recombination environmentally driven of plant holobiont induces defensive metabolic reprogramming to maintain the balance of endophytic bacteria interests to affect the quality of medicinal materials .This project designs four deficiency and two kinds of climate stress combination model. Base on the root selection culture, layered combinations are carried out from endophytic fungi, bacteria, actinomyces and to pathogenic bacteria. The Illumina and LC-MS and ICP-MS technology are adopted to determine the bacteria abundance and metabolism , and to sudy the relationship of stress, all creatures, physiological metabolism ,rhizosphere effect and the effective components . The holobiont responsed mechanism of the environment-driven quality changes of medicinal materials is to be revealed to find the new insights on the quality formation mechanism of traditional Chinese medicine, and the new ideas and methods for the ecological cultivation of medicinal plants are to be put forward.
