剥皮性是猕猴桃育种的重要目标之一,但目前研究较少。毛花猕猴桃(Actinidia eriantha Benth)是我国特有的野生种质,其果实成熟后具有易剥皮的独特特征。本项目拟选用江西境内发掘的两个果实剥皮表现为难(‘54号’)与易(‘78号’),自然地理位置分布较近且遗传差异小(遗传相似系数0.75)的毛花猕猴桃单株为试材,采集其成熟前后的果实,观测剥皮性状及果皮解剖结构;同时通过代谢组和转录组测序,筛选出两者间差异代谢物和富集通路上的差异表达基因。再利用200份(已收集118份)野生毛花猕猴桃单株进行差异表达基因关联分析,获得与剥皮性状关联具显著效应的候选基因,经Q-PCR、原核表达、转基因等技术对其进行功能验证。本项目旨在明确毛花猕猴桃果实剥皮性状形成相关的生理生化机制和分子机理,为毛花猕猴桃野生资源的开发利用,商业化猕猴桃品种剥皮性状的改良,以及猕猴桃新品种的选育推广提供依据。
英文摘要
Though fruit peeling is one of the important targets of kiwifruit breeding, there were few reports on it until now. Fruit of kiwifruit Actinidia eriantha which is a unique wild germplasm in China can develop the ability for peel detachment when they are ripe and soft. The fruit of a A. eriantha line (‘78’) with good peeling and another line (‘54’) with poor peeling will be selected as the materials for this research, as their geographic location was near and genetic similarity coefficient was high (0.75). During the development of peel detachability, the structure of the skin of fruit will be observed and the metabolomics as well as the transcriptomics of the outer pericarp of fruit will be tested and analyzed. After comparing the results of the metabolomics and transcriptomics, the differentially expressed genes involved in the synthesized pathway of main changing metabolites will be screened out and then will be conducted the association analysis in two hundreds natural A. eriantha lines with different peel detachments. The functions of the candidate genes which would show significant effects after association analysis will be assayed and identified by the Q-PCR technique, prokaryotic expression system and tomato transformation systems. This study aims not only to make clear the molecular mechanism of peel detachability of A. eriantha, but also to provide theoretical basis for the utilization of wild resources of A. eriantha and the improvement of the fruit peeling of commercial kiwifruit cultivars.
