海水鱼配合饲料过度依赖富含n-3HUFA的鱼油是制约其养殖业发展的重要因素,提高鱼体对n-3HUFA的摄取有助于减少饲料中鱼油的添加水平。近年来的研究显示,脂肪酸结合蛋白(FABP)与脂肪酸含量密切相关。申请人前期在海水鱼卵形鲳鲹中发现,随着其肝脏n-3HUFA含量的增加,FABP4及cAMP/PKA/PPARγ通路相关基因表达也上调,且FABP4启动子含有转录因子PPARγ的结合位点。提示FABP4可能参与肝脏对n-3HUFA的摄取,且该作用可能受cAMP/PKA/PPARγ通路调控。为此,本项目拟在卵形鲳鲹肝细胞上采用基因过表达、RNAi、WB、GC-MS等手段先确认FABP4在肝细胞n-3HUFA摄取中的作用,再弄清该作用是否通过cAMP/PKA/PPARγ通路对其基因的转录调控而实现。研究结果将丰富鱼类营养学理论知识,为降低海水鱼配合饲料中鱼油的添加水平、节约海洋资源提供新的思路。
英文摘要
The excessive dependence of marine fish feed on fish oil rich in n-3 HUFA is an important factor restricting the development of its aquaculture, increasing the uptake of dietary n-3 HUFA is contribute to reducing the fish oil level in feed. Recently, studies have shown that the fatty acid-binding protein (FABP) is closely related to the fatty acid content. Previously, the applicant found that with the increasing of experimental Trachinotus ovatus liver n-3HUFA content, the expression of FABP4 and cAMP/PKA/PPARγ pathway related genes was up-regulated, and several binding sites of transcription factor PPARγ were predicted in FABP4 promoter. These results suggest that FABP4 may be involved in T. ovatus liver uptake of n-3 HUFA, and this effect may be regulated by the cAMP/PKA/PPAR pathway. Therefore, this research tries to use gene overexpression, RNAi、WB、GC-MS on the T. ovatus liver cells to firstly confirm the role of FABP4 in the uptake of n-3 HUFA of liver cells, and then find out whether this role is realized through its transcriptional regulation by the cAMP/PKA/PPAR pathway. The results will enrich the theoretical knowledge of fish nutrition and provide a new idea for reducing the fish oil level in marine fish feed and saving marine resources.
