铁元素与其贮存蛋白-铁蛋白(Ferritin)对于生物体生命过程意义重大。最近的研究表明,家蚕铁蛋白在应答病原入侵时参与家蚕免疫调节过程,但其机制尚缺乏深入研究。本课题组前期研究发现,在家蚕核型多角体病毒(BmNPV)感染后的不同抗性家蚕材料中,Ferritin及相关基因表达变化明显,且Ferritin与BmNPV存在互作现象,提示宿主Ferritin参与了病原入侵的应答。本项目基于前期研究基础,拟通过进一步筛选与家蚕Ferritin互作的NPV表面蛋白,明确Ferritin及铁离子代谢对NPV入侵的影响;分析Ferritin在NPV入侵过程中对ROS聚集的抑制,以及Ferritin对JNK激活的抑制和对细胞凋亡的影响,从而阐明Ferritin参与NPV入侵宿主的调控途径;明确Ferritin与NPV互作对致病性影响的结果,为进一步解析家蚕抵抗NPV入侵的机理提供依据。
英文摘要
Iron and its storage protein-ferritin are of great significance to organisms. Recent studies have shown that silkworm ferritin participates in the immune regulation of silkworm in response to pathogenic invasion, but the mechanism is still unclear. Our previous studies found that the expression of ferritin and related genes changed significantly in different resistant silkworm strains infected with nuclear polyhedrosis virus (BmNPV), and there was interaction between Ferritin and BmNPV, suggesting that the host Ferritin participated in the response of pathogen invasion. Based on previous work, this project intends to further screen NPV surface proteins interacting with silkworm Ferritin, clarify the effects of Ferritin and iron metabolism on NPV invasion, analyze the inhibition of Ferritin on ROS aggregation during NPV invasion, and the inhibition of Ferritin on JNK activation and apoptosis, so as to clarify the regulatory pathway of Ferritin in NPV invasion. The results analyze during the interaction between Ferritin and NPV on pathogenicity will provide basis for further understanding the mechanism of resistance of silkworm to NPV invasion.
