内向整流钾离子(Kir)通道在动物体内血液循环,神经传导,激素分泌中发挥重要作用。昆虫Kir通道的典型结构特征和高等动物的相似,但其药理学性质存在显著差异。因此以昆虫Kir通道为靶标开发高效安全杀虫剂具有巨大潜力。但昆虫Kir通道研究较少,目前尚没有弄清其天然状态下的分子组成,一些关键的必需组成亚基有待分离鉴定。本项目拟以小菜蛾为研究材料,通过现代分子生物学技术克隆Kir通道的必需亚基,确定其天然分子组成,结合细胞生物学技术在HEK-293细胞中表达有生物学功能的Kir通道,利用药物结合实验和膜片钳技术研究Kir通道的药物结合位点和选择性机理,建立Kir通道与抑制剂(激活剂)相互作用分子模型,为高效安全杀虫剂的分子设计和开发提供新靶标、新思路和新途径。
英文摘要
Inward rectifier potassium ion (Kir) channels play an important role in animal blood circulation, nerve conduction and hormone secretion. The typical structural characteristics of insect Kir channels are similar to those of higher animals, but their pharmacological properties are significantly different. It has a potential to develop efficient and safe insecticides targeting insect Kir channels. However, little research has been done on insect Kir channels. At present, the molecular composition of insect Kir channels in its natural state has not been clarified. Some key constituent subunits need to be isolated and identified. In this study, diamondback moth (Plutella xylostella) was used as material to clone the necessary subunits of Kir channels by modern molecular biology technology, determine its natural molecular composition, express Kir channels with biological function in HEK-293 cells by cell biology technology, study drug binding sites and selective mechanism of Kir channel by drug binding experiment and patch clamp technology, and establish interaction models of Kir channel and inhibitor (stimulation). It will provide new targets, new ideas and new ways for the development of efficient and safe pesticides.
