水稻抽穗期是决定品种地区与季节适应性的重要农艺性状,我们前期通过筛选水稻抽穗期的突变体,发现ehd5突变体的抽穗期在长短日照下相对于野生型均推迟一周左右。通过Mutmap方法定位克隆到了EHD5基因,发现其编码一个RNA结合蛋白,利用转基因互补和CRISPR敲除对该基因进行了功能验证,确认该基因就是EHD5。本项目拟在前期基因克隆的基础上进一步明确EHD5基因表达模式,蛋白亚细胞定位、RNA结合活性等基本特性,并通过多种组学手段(RNA-seq、PAS-seq、CLIP-seq)在全基因组水平上鉴定EHD5所结合的靶RNA,分析其结合的核心序列,并通过RNA EMSA,RIP-qRT-PCR等手段进行验证,进一步通过构建双突变体等手段验证其遗传关系,阐明EHD5调控抽穗期的信号途径。鉴于EHD5对抽穗期的调控不受光周期的影响,本项目还将评估EHD5基因在水稻抽穗期育种中的利用价值。

The heading date of rice is an important agronomic trait that determines the adaptability of varieties to regions and seasons. We screened for rice heading date mutants and found that the heading date of ehd5 mutant was delayed by about one week compare with wild type under both the long and short daylength. The EHD5 gene was cloned by the Mutmap method and found to encode an RNA-binding protein. The candidate gene was further verified by transgene complementation and CRISPR knockout, and was confirmed to be EHD5. Based on the previous gene cloning, this project aims to further clarify the basic characteristics of EHD5, including gene expression pattern, protein subcellular localization and RNA binding activity. Further, we will use multiple omics methods (RNA-seq, PAS-seq, CLIP-seq) to identify the target RNA bound by EHD5, and the binding core sequence will also be isolated. The target will be further verified by RNA EMSA, RIP-qRT-PCR methods. Further, the genetic relationship of EHD5 and its target genes will be analyzed by constructing double mutants. This study will clarify the signal pathway the EHD5 controlling heading date. Since EHD5 can regulating heading date not affected by the daylength, it is a good target for rice heading date improvement, this project will also assess its value on breeding.
