马铃薯甲虫是全球马铃薯主产区的毁灭性害虫,其强大的繁殖力,导致种群爆发式增长,疫情发生面积持续扩大,自该虫传入我国以来,严重威胁我国的粮食安全,因此,补足该虫生殖基础研究短板,寻找绿色防控策略和高效、专一的防治靶标成为当前亟待解决的科学问题。卵黄发生是昆虫卵发育的基础,研究表明海藻糖参与卵黄原物质积累,调控卵黄发生过程,但作用机理不详。我们前期研究发现马铃薯甲虫雌虫海藻糖含量与保幼激素滴度,类胰岛素通路基因表达及卵黄发生正相关。推测海藻糖通过物质代谢和激素信号,协同调控马铃薯甲虫的卵黄发生过程。本项目拟在已有基础上,采用RNAi、免疫印迹、HPLC等实验技术,通过活体基因敲减和离体孵育等研究手段,探讨激素信号响应海藻糖含量变化的信号转导通路,及调控卵黄发生的互作关系,从而揭示海藻糖调控马铃薯甲虫卵黄发生的分子机理。研究结果可弥补马铃薯甲虫生殖生理基础研究的不足,为马铃薯甲虫防控提供靶标。
英文摘要
The Colorado potato beetle, Leptinotarsa decemlineata is a notorious pest on potato crops worldwide. Its strong fecundity has led to explosive population growth and a continued expansion of the outbreak area. China was first recorded on the pest in Ili River Valley area in 1993. Since then, it spread rapidly to all over the southern region of Tianshan Mountain and causing huge potato yield losses every in Xinjiang autonomous region. The beetle continues to spread from west to east, and the risk of spreading to other provinces is huge, which seriously threatens the food security of China. Therefore, it is an urgent scientific problem to make up for the deficiency of basic research on reproduction while searching for safety control strategies and effective target molecules for the beetles control. Vitellogenesis is the basis of insect egg development. Studies have shown that trehalose participates in the accumulation of vitellogenin and regulates the process of vitellogenesis, but the underlying mechanism is unknown. Our previous study found that the trehalose content of the female was positively correlated with juvenile hormone titer, insulin-like pathway gene expression and vitellogenesis. It is suggested that trehalose regulates the vitellogenesis through nutrient metabolism and hormone signals in L.decemlineata. In this project, RNAi, immunoblotting, LC-MS, HPLC and other experimental techniques are used to explore the molecular pathway of hormone signal sensing trehalose level, as well as the interaction of various hormone signals regulating vitellogenesis, thus revealing the molecular mechanism of trehalose regulating vitellogenesis of L.decemlineata. The results can fill in the deficiencies of basic research on reproduction, and provide target genes for Colorado potato beetle control.
