既往研究证实小梁网能调节房水流出阻力,且和自主神经相关。同时小梁网细胞间连接是小梁网调节房水流出阻力的重要靶点。探索自主神经对小梁网细胞间连接的调控,可能是揭示小梁网途径房水流出阻力变化的关键。我们研究发现:自主神经兴奋可扩张小梁网,降低眼压;小梁网局部存在自主神经表达;青光眼模型鼠小梁网细胞间连接蛋白表达、小梁网细胞骨架和眼压均异常;局部给与自主神经工具药后,青光眼模型鼠小梁网细胞间连接蛋白表达、小梁网细胞骨架和眼压恢复正常;由此我们假设:自主神经可通过调节小梁网细胞间连接,改变小梁网途径房水流出阻力。本项目拟用免疫荧光、免疫印迹、电子显微镜等,观察青光眼模型鼠局部运用工具药后小梁网细胞间连接、小梁网开放状态和房水流出速率变化,以及观察压力刺激、工具药干预下,体外培养小梁网细胞的细胞间连接和细胞间水通透性变化。为自主神经通过小梁网细胞间连接调节房水外流研究和青光眼治疗提供新思路。

Previous studies have indicated that the trabecular meshwork (TM) could regulate the aqueous humor outflow resistance and this regulation was associated with autonomic nerves. Moreover, the intercellular junction of TM cell was a major target in the regulation of aqueous humor outflow resistance. Investigating the regulation effect of autonomic nerves on the intercellular junction of TM cell could be the key point to reveal the mechanism for the aqueous humor outflow resistance alteration. Our previous studies have found that, the activation of autonomic nerves expanded TM and reduced intraocular pressure (IOP); autonomic nerves and transmitters could be found in the TM; the expression of intercellular junction protein of TM, the cytoskeleton of TM and IOP were abnormal in the glaucomatous rat model. Furthermore, after the local application of autonomic nervous drugs, the expression of intercellular junction protein of TM, the cytoskeleton of TM and IOP returned to normal in the glaucomatous rat model. Thus, we hypothesized that the autonomic nerves could alter the aqueous humor outflow resistance via regulating the intercellular junction of TM cell. In this present study, using immunofluorescence staining, western blotting and electron microscope, we prepared to observe the changes in the intercellular junction of TM, the expansion state of TM and aqueous humor outflow rate in glaucomatous rat model after the local application of autonomic nervous drugs, and to observe the changes in the intercellular junction and intercellular liquid permeability of cultured TM cell after the high-pressure or autonomic nervous drugs stimulation, hoping to provide novel insights for the regulation effect of autonomic nerves on the aqueous humor outflow via TM intercellular junction modification and for the anti-glaucoma treatment in clinic.
