远处转移是乳腺癌死亡的主要原因,转移新途径的探索是肿瘤研究领域的核心科学问题。申请人在长期研究细胞膜及骨架蛋白的基础上发现乳腺癌中存在一种由质膜构成的中空管状结构(Tube-like vessel),肿瘤细胞包裹其中且与乳腺癌远处转移显著相关(Nature, submitted),测序发现ESR1,MAP3K1和PIK3R1与其生成相关,我们推测Tube-like vessel可能是乳腺癌远处转移的独立途径。本研究拟首先通过体外实验模拟Tube-like vessel的生成和肿瘤细胞的转移,然后利用高通量测序、蛋白质组学等手段探索调控Tube-like vessel的驱动基因,再通过动物模型研究Tube-like vessel的转移机理和阻断途径,最后前瞻性临床试验验证此科学假说。本研究将初步证实Tube-like vessel新的肿瘤转移途径,为乳腺癌转移理论及治疗策略提供新思路。

Distant metastasis is the main cause of death of breast cancer, and the exploration of new ways of metastasis is the core scientific problem in the field of oncology research. In the previous work, we found that there was an extracellular membrane compartment (Tube-like vessel) composed of plasma membrane in breast cancer, which was encapsulated by tumor cells and significantly related to distant metastasis of breast cancer (Nature, submitted). RNA-seq showed that ESR1,MAP3K1 and PIK3R1 were related to the production of Tube-like vessel, so we speculated that Tube-like vessel may be an independent pathway of distant metastasis of breast cancer. In this study, we first simulate the formation of Tube-like vessel and metastasis of tumor cells in vitro, then explore the driving genes that regulate Tube-like vessel by means of high-throughput sequencing and proteomics, and then establish an animal model of tumor metastasis to study the metastasis mechanism and blocking pathway of Tube-like vessel. Finally, prospective clinical trials are used to verify this scientific hypothesis. This study will initially confirm the existence of a new tumor metastasis pathway of Tube-like vessel, and provide new ideas for the improvement of breast cancer metastasis theory and treatment strategy.
远处转移是乳腺癌相关死亡的主要原因,然而高侵袭性乳腺癌细胞转移的早期过程和分子机制尚未完全清楚,且缺乏特异性靶向干预策略。申请人在乳腺癌干细胞与肿瘤器官转移领域的长期研究积累(Elife,2021;Mol Cancer,2022;Drug Resist Updat,2023),为乳腺癌相关治疗提供了新理论和新靶点。本项目预实验通过人工智能分析技术对发生转移的乳腺癌患者原发灶进行特征识别,首次发现与转移相关的高信号区,并利用蛋白质组学和组织多色成像技术鉴定到隧道样新结构。进一步利用发生转移的乳腺癌患者原代肿瘤细胞进行三维培养,在体外三维模型中鉴定到促进癌细胞在三维环境中快速迁移的新通道——隧道样迁移轨迹(Tunnel-like migration trajectory,TMT),其结构和功能特征区别于已知的血管、淋巴管和血管拟态等转移相关结构。本项目拟通过多维度研究系统解析TMT的形成机制及其在乳腺癌转移中的作用。首先,利用活细胞实时成像技术动态追踪体外三维培养模型中TMT的形成过程,结合超分辨荧光成像与原位电镜技术鉴定TMT的显微特征。其次,采用显微捕获技术富集三维培养模型中的TMT,联合蛋白质组学和多维染色技术解析其组成成分,进一步阐明TMT形成细胞的表型特征及其分子调控网络并加以验证。在此基础上,利用PDX和CDX动物模型验证TMT的生物学功能,并通过782例乳腺癌原发灶样本染色和回顾性分析,明确TMT与乳腺癌转移等不良预后的相关性。最后,借助体内外模型验证调控乳腺癌干细胞形成TMT的关键影像因素,并研发靶向干预方法。综上,本研究拟通过多组学与动态成像技术,系统解析乳腺癌原发灶中TMT的形成过程、组成成分、形成机制及生物学功能,揭示GJB3是调控乳腺癌干细胞TMT形成的关键因子,为乳腺癌转移的早期机制研究提供新视角,有望为预测转移发生风险及开发靶向精准疗法提供理论依据。(Cell Research杂志修回)
