环境中纳米塑料的广泛存在严重威胁人类健康,但对其毒性效应尚缺乏深入研究。本项目拟以聚苯乙烯纳米塑料为代表,利用RNA干扰、免疫组化、免疫荧光、透射电镜、电感耦合等离子体质谱等技术,研究纳米塑料在小鼠体内的转运吸收机制和毒性效应。通过纳米塑料暴露小鼠和荧光纳米塑料暴露肠上皮细胞实验,明确纳米塑料在小鼠体内转运吸收与外排的分子机制;基于纳米塑料暴露小鼠实验,检测小鼠长期暴露纳米塑料后肠黏膜组织完整性、氧化应激和免疫功能的变化,研究纳米塑料对小鼠肠黏膜屏障功能的毒性效应;通过金核-聚苯乙烯塑料外壳纳米颗粒暴露小鼠实验,获得小鼠多种组织中纳米塑料浓度,基于毒理毒代动力学/毒效动力学模型,利用三参数Weibull阈值模型预测其风险阈值,进而推算人体健康风险阈值。本项目研究结果不仅能够阐明纳米塑料长期暴露后在小鼠体内的转运吸收机制和毒性效应,还能为探索纳米塑料对人体健康的影响提供实验依据。

The widespread existence of nanoplastics in the environment is a serious threat to human health, and the toxic effect of nanoplastics is still lack of in-depth study. In this project, polystyrene nanoplastics will be used as a representative, and the transportation, absorption and toxic effects of nanoplastics in mice are studied using RNA interference, immunohistochemistry, transmission electron microscopy, immunofluorescence and inductively coupled plasma mass spectrometry (ICP-MS). The absorption, transportation and efflux mechanism of nanoplastics in mice are studied by exposing mice and intestinal epithelial cells with nanoplastics; After long-term exposure of nanoplastics in mice, the changes of intestinal mucosal tissue integrity, oxidative stress and immune function are studied, which can reveal the toxic effects of nanoplastics on intestinal mucosal barrier function; The concentrations of nanoplastics in various tissues of mice are obtained in mice exposed by nanoparticles with Au core and polystyrene plastic shell. Based on the toxicity-based toxicokinetic/toxicodynamic (TBTK/TD) and the three-parameter Weibull model, the risk threshold of nanoplastics is predicted in mice. Furthermore, the risk threshold of nanoplastics in human is evaluated according to risk threshold of mice. The research results of this project will clarify the mechanism of polystyrene nanoplastics transportation and absorption and toxic effects of nanoplastics in mice after long-term exposure, and provide experimental basis for exploring the impact of nanoplastics on human health.
纳米塑料污染是全球环境科学关注的热点问题,其对生物体的潜在毒性及健康风险亟需深入研究。本项目以聚苯乙烯纳米塑料(PS-NPs)为研究对象,系统探讨了其在体内的吸收、转运机制及毒性效应,取得了系列研究结果。通过构建不同浓度(0.1、1、10mg/L)纳米塑料暴露的小鼠模型,结合透射电镜、免疫组织化学和荧光标记技术,研究发现PS-NPs主要通过caveolin和clathrin介导的内吞途径进入细胞,并通过Rab7、LAMP1等内吞相关蛋白的协同作用完成跨上皮转运,同时激活自噬途径调控其胞内分布,明确了PS-NPs的生物吸收和转运机制。长期暴露于PS-NPs的小鼠肠黏膜屏障功能显著受损,包括肠绒毛缩短、隐窝深度降低以及紧密连接蛋白(occludin、claudin-1、ZO-1)表达下调,伴随显著的氧化应激反应(MDA升高,SOD和GSH-Px活性下降)及炎症反应(IL-6、TNF-α上调)。同时,肠道免疫功能紊乱表现为B淋巴细胞比例升高,固有层T细胞中γδ⁺CD8⁺T细胞显著减少,提示PS-NPs可能通过破坏免疫调控加剧健康风险。本研究构建了Au核标记的PS-NPs暴露小鼠模型,通过ICP-MS技术定量分析其在肠道和肝脏等组织中的分布,并结合毒理毒代动力学/毒效动力学(TBTK/TD)模型,量化评估了其剂量-效应关系,结果显示PS-NPs在体内的分布呈显著的剂量和时间依赖性。结合三参数Weibull模型的外推计算,预测了100nm PS-NPs对人体健康的风险阈值浓度为0.07554ng/kg,颗粒数为952.9×10³个/kg,为评估纳米塑料暴露的健康风险提供了科学依据。本项目成果揭示了PS-NPs的分子转运机制和毒性效应,已发表多篇高水平研究论文,并在国内学术会议上作特邀报告,对微塑料污染治理与公共健康保护提供了重要理论支持。
