缺氧是诱导胃癌细胞发生EMT和转移的重要因素。我们前期发现circIREB2在伴淋巴结转移的胃癌组织和缺氧胃癌细胞中高表达,并参与缺氧诱导的胃癌EMT。进一步研究发现circIREB2具有编码多肽的潜能。目前对于circIREB2在缺氧诱导胃癌EMT和转移中的具体作用及机制尚不清楚。基于此,我们提出缺氧诱导circIREB2及其编码多肽的表达上调,经Wnt/β-catenin通路促进胃癌EMT和转移的假设。因此,拟从三个层面进行研究:1、circIREB2在胃癌EMT和转移中的作用;2、circIREB2如何通过编码多肽并参与胃癌EMT和转移的机制;3、circIREB2及其编码多肽作为胃癌治疗靶点和预后指标的潜能。本研究旨在揭示circIREB2在缺氧诱导胃癌EMT和转移中的作用及其机制,为胃癌转移机制提供新的理论依据,亦可为胃癌诊治提供新的潜在靶点,具有重要理论意义和潜在临床应用价值。
英文摘要
Hypoxia is an important factor inducing EMT and metastasis in gastric cancer cells. Our previous research found that circIREB2 which was highly expressed in gastric cancer tissues with lymph node metastasis and hypoxic gastric cancer cells, had been involved in hypoxia-induced gastric cancer EMT. It was also found that circIREB2 had the potency to encode a polypeptide in our further research. The specific role and mechanism of circIREB2 in hypoxia-induced gastric cancer EMT and metastasis remains unknown. We hence propose that hypoxia induces increased expression of circIREB2 and its encoded polypeptide, and promotes EMT and metastasis of gastric cancer via the Wnt/β-catenin pathway. Therefore, we will proceed with our research from the following three layers. 1. The role of circIREB2 in gastric cancer cell EMT and metastasis; 2. How circIREB2 encodes peptides and participates in the mechanism of gastric cancer EMT and metastasis; 3. The potential of circIREB2 and its encoded polypeptides as targets and prognostic indicators of gastric cancer. With significant theoretical and clinical importance, this study which aims to reveal the role and mechanism of circIREB2 in hypoxia-induced gastric cancer EMT and metastasis, will provide new theoretical basis for gastric cancer metastasis, and provide new targets for the prevention and treatment of gastric cancer.
胃癌是常见的消化道肿瘤。已有研究发现缺氧是胃癌进展的关键诱因。然而针对pre-mRNA通过剪接体生成成熟mRNA过程中伴随产生的大量circRNA在缺氧诱导胃癌进展中的作用还认识有限。本研究旨在探讨circIREB2通过编码蛋白IREB2-268aa参与胃癌进展的功能和机制研究,以期为胃癌诊治提供新的诊治靶点。1、我们结合circRNA芯片结果、编码蛋白能力预测以及表达验证结果,发现circIREB2在胃癌组织和缺氧培养胃癌细胞中显著高表达;同时circIREB2包含IRES翻译起始元件和ORF序列,具备翻译潜能。2、我们结合circIREB2序列分析和验证翻译潜能的相关实验结果,发现circIREB2中的IRES序列具有翻译起始活性且ORF序列可以被翻译;circIREB2所翻译的IREB2-268aa含有23个氨基酸组成的特异序列,可以被制备的多克隆抗体识别并验证。3、我们结合细胞定位预测和实验验证结果,发现IREB2-268aa主要定位于线粒体内膜,且在胃癌组织和缺氧培养的胃癌细胞中高表达。同时结合铁死亡相关实验、以及体外和体内功能实验结果,我们发现IREB2-268aa可以通过降低线粒体Fe2+和线粒体膜去极化来减少脂质过氧化物堆积,进而抵抗铁死亡以增强胃癌增殖和转移能力,促进胃癌进展。4、我们结合RNA-seq、蛋白互作实验、免疫共定位实验和蛋白稳定性实验结果,发现IREB2-268aa和DHODH共定位于线粒体内膜,且IREB2-268aa可以结合DHODH并增强DHODH蛋白稳定性。5、我们结合体外实验和体内实验,进一步发现IREB2-268aa联合DHODH抑制剂是胃癌患者(特别是GPX4low胃癌患者)的潜在治疗靶点。项目相关研究成果在Journal of Translational Medicine等SCI期刊发表论文11篇。
