刺参是我国重要海洋经济物种,产业规模不断扩大,其基础生物学研究受到关注。刺参于初夏完成生殖活动,继而进入“夏眠”,严重影响养殖生产效率。本项目围绕刺参生殖与夏眠生理状态转变这一特殊阶段,在前期刺参G蛋白偶联受体基因预测及多组织转录表达差异分析、相关神经激素信号系统预测与鉴定、刺参成体组织细胞培养方法优化等工作基础上,从神经内分泌研究角度,运用代谢组学、多肽组学及转录组学等高通量测试分析手段,结合分子及细胞生物学和生理功能研究方法,查明刺参生殖与夏眠生理状态转变过程中的神经内分泌调控特征,阐明神经激素与受体的作用活性及介导的细胞信号转导特征,解明关键神经内分泌信号系统的调控关系及其生理功能,综合构建神经内分泌调控网络模型,探明刺参生殖与夏眠状态转变的神经内分泌调控机制。本项目的实施,将从本质上揭示刺参生殖与夏眠转变的内在调控机理,丰富刺参基础生物学研究理论,获得动物生殖与休眠调控研究新认识。
英文摘要
Sea cucumber Apostichopus japonicus is an important marine economic species in China with expanding industrial scale, and a great number of researches are focusing on its basic biological processes. A. japonicus ceases reproductive activities in early summer, and then enters into the "aestivation" status, which seriously affected the production efficiency of aquaculture. Based on obtained results, from the previous studies, the prediction and transcriptional analysis of A. japonicus G protein coupled receptors, the prediction and identification of neurohormonal systems, and the optimization of primary cell culture and tissue culture techniques, this project focuses on the special stage between reproduction and aestivation of A. japonicus from the perspective of neuroendocrine research. Using high-throughput detection and analysis, such as metabolomics, peptidomics and transcriptomics, combined with molecular, cellular and physiological researches, we aim to clarify neuroendocrine regulatory characteristics during the transiting stage, evaluate the interaction between neurohormones and receptors, clarify the characteristics of cell signal transduction, understand the regulatory relationship and physiological functions of key neuroendocrine signal systems, and comprehensively construct the neuroendocrine regulatory network and explore the regulatory mechanism of this model. The implementation of this project will essentially reveal the internal regulatory mechanism of the physiological transition stage in sea cucumber and gain a new understanding of animal reproduction and dormancy control.
刺参 (Apostichopus japonicus) 具有显著的季节性繁殖与夏眠等生理特征,这些状态的转变受神经内分泌系统的精密调控,然而有关动物生殖与夏眠的调控关系及机制研究仍然较少。本项目以刺参为研究对象,系统解析了刺参生殖与夏眠生理状态转变的神经内分泌调控特征与机制,取得了以下主要成果:①神经肽与G蛋白偶联受体(GPCRs)鉴定与筛选:共鉴定了33个神经肽前体和521个GPCRs,初步明确了其时空表达特征;筛选出与生殖和夏眠显著相关的神经因子及GPCRs。②5-羟色胺(5-HT)系统功能研究:发现外源性5-HT显著增强刺参的运动行为与触手摄食频率,同时促进肠道消化酶活性、稳定葡萄糖水平和激活糖酵解通路;进一步鉴定了刺参的5-HT受体,揭示了5-HT信号系统能量代谢和行为中的核心作用。③雌激素与褪黑素系统协同参与生殖调控研究:鉴定了刺参的雌激素受体、褪黑素受体和褪黑素合成酶,揭示了两大内分泌系统在雌性生殖调控中的协同作用:褪黑素通过其受体激活雌激素分泌,进一步通过受体活化调控刺参雌性性腺发育。④性腺发育调控的转录组学解析:通过比较转录组分析,探讨了刺参雌雄性腺发育过程中关键基因(如Foxl2、CYP2B6、Dmrtb1和SYCP2等)在性腺发育调控中的作用机理。⑤非编码RNA在肠道退化中的调控机制:预测并构建了lncRNA-miRNA-mRNA调控网络,发现非编码RNA通过调控关键基因(如AjSOX9和AjWNT9B),参与刺参肠道退化与能量代谢适应过程调控。⑥核心神经内分泌调控网络的构建:通过蛋白互作网络(PPI)分析,明确了AjTRHP和AjCRZP等神经肽在能量平衡和繁殖调控中的核心地位,识别出AjGALR2和AjCHRM5等枢纽基因,初步构建了刺参神经内分泌调控网络。本项目成果发表SCI论文7篇、英文专著章节1篇,培养研究生9名(其中6名硕士已毕业)。项目系统揭示了刺参生殖与夏眠状态转变的神经内分泌调控机制,为理解无脊椎动物神经内分泌系统的分子调控提供了新认知。
