英文摘要
Inverse Leidenfrost effect generally happens in the process of vitrifying biosamples by quenching with liquid nitrogen, which has an adverse effect on the qualities of cryopreserved biosamples. The current proposal presents a research idea that the synergism between inverse Leidenfrost effect and vitrification behaviors of microscale biosamples while quenching in liquid nitrogen is a kind of complicated heat and mass transfer problem in nature. And it is necessary to assess the effect of inverse Leidenfrost on the vitrification trend and find out an approach to avoid this adverse effect for biosamples long-term storage in low temperature. In current proposal, three critical issues will be mainly concerned such as how to develop the quantitative assessment method for both vitrification degree inside cryopreserved biosamples and key parameters of inverse Leidenfrost phenomenon, how to accurately characterize the synergism mechanism between inverse Leidenfrost effect and vitrification behaviors of biosamples by quenching, and how to eliminate the adverse effect of inverse Leidenfrost effect experimentally for the purpose of cryopreservation application. Importantly, the current proposal aims to figure out the key scientific issues related to vitrifying microscale biosamples by quenching. This should be very helpful for developing high-throughput, high-quality, and long-term storage protocols of important biological resources in China. Also, it can meet the demands in some basic scientific field and pharmaceutical industry.
本项目提出微小尺度生物样品淬冷玻璃化行为与逆Leidenfrost效应是具有协同作用的复杂传热传质问题的研究思想,探究逆Leidenfrost效应对微小尺度生物样品玻璃化形成能力的多种影响因素。从以下7个方面进行了研究:(1)典型低温保护剂溶液体系热物性参数和渗透参数分析及Raman光谱数据库建立;(2)基于Raman光谱获得样品内部结晶特性研究;(3)液氮淬冷过程中逆Leidenfrost现象气膜消涨可视化实验研究与理论分析;(4)微小尺度生物样品淬冷玻璃化冻存前保护剂加载工艺研究;(5)微小尺度生物样品负压蒸发预脱水过程的细胞损伤评估与玻璃化保存研究;(6)基于低频振动调控冻存载体池沸腾传热特性研究;(7)基于表面改性低导热涂层方法调控冻存载体沸腾传热特性研究。该项目利用多种调控手段解决了微小尺度生物样品玻璃化保存过程中逆Leidenfrost效应的关键问题,并拓展了蒸发预脱水冻存技术及Raman光谱低温保存全流程评估等多个前瞻性研究课题,对于开发创新的低温保存方法奠定了重要研究基础。.通过本项目的实施,共发表了各类论文31篇,其中SCI检索26篇(中科院二区以上论文18篇),获得国家专利授权10项,其中发明专利7项,制定并解读团体标准1项,获上海市科学技术二等奖1项。项目共计培养7名博士,9名硕士。项目团队主办3次全国性学术论坛,11人次参加国内外重要学术会议。
