早期断奶引发氧化应激是诱发“断奶应激综合症”的重要原因,Nrf2/AREt是维持细胞内氧化还原平衡、抗氧化应激的重要通路,植物精油中活性成分激活Nrf2依赖的适应性应答是其提高机体抗氧化能力的有效调控手段。牛至精油是由牛至提取的植物精油,抗氧化潜力为96.6-137.5mmol。然而,关于牛至精油缓解早期断奶犊牛氧化应激作用机理尚不明确,关于牛至精油调控Nrf2/ARE通路缓解早期断奶犊牛肠道氧化损伤的研究未见报道。本研究首先以早期断奶犊牛体内氧化应激模型为研究对象,初步探索牛至精油缓解早期断奶犊牛氧化应激的作用和机理;紧接着以早期断奶犊牛体外模型为研究对象,通过isRNA干扰技术挖掘牛至精油对缓解早期断奶犊牛氧化应激的分子调控机制,探究牛至精油可否通过调控Nrf2/ARE通路缓解氧化应激;最后,通过体内和体外模型试验结果的相互验证,进一步揭示牛至精油缓解早期断奶犊牛氧化应激的调控机制。

Oxidative stress caused by early weaning is an important cause of “weaning stress syndrome”. Nrf2/ARE signaling pathway is a system that converts exogenous stimuli into eukaryotic transcriptional response, and is an important pathway to maintain intracellular REDOX balance and antioxidant stress. Active ingredients in essential oils activate Nrf2-dependent adaptive responses, which is considered to be an effective regulatory means for plant extracts to improve the body's antioxidant capacity. Oregano essential oil is a plant essential oil extracted from oregano with antioxidant potential of 96.6-137.5mmol. However, the mechanism of oregano oil to relieve oxidative stress in early weaned calves is still unclear and studies on whether oregano oil can alleviate intestinal oxidative damage in early weaning calves by regulating the Nrf2/ARE signaling pathway have not been reported. In this study, the in vivo model of early weaned calves was firstly used as the research object to preliminarily explore the effect and mechanism of oregano essential oil to relieve the oxidative stress of early weaned calves. Secondly, the in vitro model of early weaning calf was taken as the research object, and the molecular regulation mechanism of oregano essential oil on relieving oxidative stress of early weaning calf was explored by using isRNA interference technology, to explore whether oregano essential oil could alleviate oxidative stress by regulating Nrf2/ARE signaling pathway. Lastly, the regulation mechanism of oregano essential oil to relieve oxidative stress in early weaned calves was further revealed through the mutual verification of in vivo and in vitro model test results.
早期断奶犊牛因氧化应激引发的健康问题严重制约畜牧业可持续发展。本项目针对犊牛断奶期氧化应激综合症,以建立氧化应激模型为基础,系统研究牛至精油的剂量效应及其与丁酸钠的协同调控机制,旨在为犊牛健康养殖提供理论与技术支撑。研究主要内容包括:(1)通过Diquat诱导构建犊牛氧化应激模型,明确其剂量效应与作用路径;(2)筛选牛至精油的最优添加量,解析其通过“微生物-代谢-免疫”轴缓解氧化应激的分子机制;(3)探究牛至精油与丁酸钠的协同效应及其对瘤胃微生物与宿主代谢的调控作用。研究结果表明:①Diquat最佳诱导剂量为8 mg/kg BW,可稳定建立犊牛氧化应激模型(ADG下降40.7%,胸围增益降低35.7%),并揭示其通过抑制抗氧化酶活性与破坏肠道屏障功能的“双重路径”。②牛至精油的最优添加量为8 g/h/d,显著提升断奶犊牛生长性能(ADG达1.37 kg/d)、抗氧化能力(SOD活性提高40.7%)及免疫功能(IgA增加25.3%),并阐明其通过富集瘤胃纤维降解菌(如Lachnospiraceae_AC2044_group)和激活Nrf2/ARE通路调控能量代谢与氧化平衡的作用机制。③丁酸钠可缩短断奶时间2.5天(血清丁酸↑18.6%),但其与牛至精油联用未显著增强协同效应,且混合组因瘤胃菌群失衡(UBA1819异常增殖)导致腹泻率升高,需持续干预以维持效果。科学意义方面,首次建立犊牛氧化应激模型,填补幼龄反刍动物相关研究空白;系统揭示牛至精油通过多靶点调控宿主-微生物互作的分子机制,为植物精油替代抗生素提供理论依据。应用层面,牛至精油与丁酸钠可分别作为抗菌剂和瘤胃发育促进剂,降低犊牛腹泻率(↓15%-20%),提升饲料转化率(牛至精油组↑9.3%),预计规模化应用年潜在经济效益超10亿元。研究结果已形成SCI论文7篇、专利3项,并在多家牧场示范推广,为犊牛抗应激管理及绿色养殖技术开发提供了重要支撑。
