古代壁画是人类文化遗产的重要组成部分,微生物退化与降解作用是威胁其长久保存和展示利用的重要因素,对于壁画微生物病害的防治一直是国内外文物保护领域未能有效解决的难题。本项目针对甘肃两处石窟寺近年连续爆发的微生物病害问题,利用分离鉴定与高通量测序等技术开展壁画微生物致病类群及群落特征分析;采用现状调查与基因测序等手段完成石窟内外节肢动物多样性、种群密度及其行为特点分析;探讨壁画病害菌与节肢动物间相互关系;通过对壁画制作材料、表面污染物、节肢动物、访客活动、洞窟形制、空气交换率及环境因子等进行关联性分析,确定诱发壁画微生物病害发生发展的关键因素和成因机制;运用宏基因组学和定量蛋白质组学技术揭示微生物降解壁画的相关分子机制。提出壁画微生物防治对策并建立防治技术体系,综合评价防治效果。研究成果可为壁画微生物病害防治提供参考依据,对于提升我国文化遗产保护的科技水平亦具有重要的现实意义。

Ancient wall paintings are an important part of human cultural heritage, while microbial deterioration and degradation are important factors that threaten their long-term preservation and exhibiting utilization. The prevention and control of mural microbial diseases are problems that have not been effectively solved in the field of cultural heritage conservation at home and abroad. This project is aimed at the microbial disease problems that have been ourbreaking in two cave temples of Gansu in recent years, the isolation and identification and high-throughput sequencing techniques are utilized to carry out the analysis of mural microbial pathogenic groups and community characteristics; use condition surveys and gene sequencing to complete the diversity of arthropods inside and outside the caves, their population density and behavior characteristics can be clarified; explore the relationship between mural disease microorganisms and arthropods; take the association analysis of murals materials, surface pollutants, arthropods, visitor activities, cave formation and air exchange rate, environmental factors etc. to clearly clarify the key factors and causes of the occurrence and development of microbial diseases in murals; apply the methods of metagenomics and quantitative proteomics to reveal the molecular mechanism of microbial degradation of wall paintings. Finally we will propose countermeasures against mural microorganisms and establish a controlling technological system to solve these microbial problems, and to evaluate control effects comprehensively. The research results can provide a reference basis for the prevention and treatment of mural microbial diseases, and also have an important practical significance for promoting the scientific and technological conservation level of cultural heritage sites in China.
本项目以石窟寺壁画微生物为研究对象,利用现状调查、高通量分析、分离培养等方法确定了致害微生物与节肢动物的空间分布特点、总体及活性微生物多样性、群落组成和结构特征,揭示了微生物暴发的环境成因及其对壁画的退化机制,并开展除菌方法筛选评估,形成了机械清除与化学杀灭相结合的除菌技术。结果表明,致害微生物和节肢动物主要分布在麦积山石窟北壁,南壁较少;致害菌暴发导致壁画细菌和真菌丰富度及多样性下降;假诺卡氏菌属、放线菌属、蛛网膜菌属为优势属;优势物节肢动物为跳啮(Psyllipsocus sp.),其夏秋两季活跃,可取食菌丝体,种群密度与壁画微生物空间分布规律相吻合。炳灵寺石窟霉变主要出现在酥油残留物和岩鸽排泄物上,假诺卡氏菌属和白色侧齿霉菌占优势。夏秋季持续性降水、高湿度和洞窟内丰富的营养源是壁画微生物暴发的重要环境成因。对34个真菌菌株生物降解能力评估发现,多数有耐盐性和耐旱性,能耐受浓度达15%的NaCl和25%的PEG-6000,表明其能适应壁画表面高盐分、低湿度条件。优势致害菌Arachnomyces bostrychodes和白色侧齿霉菌生物降解范围广,对纤维素、明胶、酪蛋白、角蛋白和碳酸钙均有较强的水解能力,有耐碱、耐旱性。A. bostrychodes最适pH为9,不产酸,但分泌褐色色素,有较强碳源氧化能力,能以麦秆、麻丝、棉花、明胶等壁画制作材料作为单一营养源生长;其基因组中大部分基因功能未知,其余以碳水化合物运输和代谢占比最高;KEGG注释CDSs得到包括糖代谢、氨基酸代谢、脂类代谢等代谢通路;通过CAZy数据库比对发现其有降解纤维素和木质素的多种酶及生成青霉素的完整途径;盐分胁迫后其蛋白差异表达明显,纤维素酶活升高。通过抑菌实验及色差和ATP生物荧光评估,2-辛基-4-异噻唑啉-3-酮(辛噻酮)抑菌效果最好,利用软刷清除和负压装置无害化处理有助于致害菌防控。项目成果对提升我国石窟寺保护的科技水平具有重要现实意义。
