英文摘要
Innate immunity is the first line of the host defense against invading pathogenic microorganisms, which is regulated by multiple genes and complex biological processes at the levels of transcription, translation, and post-translation. Investigating the underlying molecular mechanisms of how innate immune signaling is precisely controlled has been one of the hottest areas in the basic research field of immunology. In this project, we will utilize Drosophila as the animal model to explore the molecular mechanisms by which RNA-binding protein Roquin plays a negative role in the STING-dependent innate immune response. The implementation of this project will not only promote our understandings of the regulatory mechanisms of STING-Ikkβ-Relish signaling pathway in Drosophila, but also deepen our knowledge of how RNA-binding proteins contribute to regulating immune response. We also hope that the research findings of this project will provide strong evidence for regulatory models of STING at the mRNA level and novel ideas for studying regulatory mechnisms of the interferon-independent STING-NF-κB signaling in mammals.
固有免疫是机体抵御外来病原微生物入侵的第一道防线,其涉及多基因多层次的转录、翻译和翻译后调控的复杂过程。探究固有免疫信号精细调控的分子机理一直是免疫生物学基础研究中的热点领域之一。本项目以果蝇为动物模型,系统深入探究了RNA结合蛋白Roquin负调控STING依赖的固有免疫反应的分子机理。项目不但揭示了Roquin通过多个功能结构域参与果蝇抗病毒固有免疫应答,而且阐明了Roquin通过结合CCR4/NOT蛋白复合体,靶向调控Sting mRNA的去腺苷化和衰败过程的分子机理。项目的实施为Sting在mRNA水平上的调控模式提供了有力佐证,并为进一步研究哺乳动物STING-NF-kB干扰素非依赖信号调控机制提供新思路。
