多型是指同一基因型响应环境预示信号诱导而产生两个或更多间断表型的现象。预示信号对未来环境的准确预测是多型维持的关键。多型生物基于环境诱导产生的表型在功能、解剖和行为上的变化远超仅由遗传引起的差异。基于多型生物的生态发育生物学研究对于发育生物学的思想和实践均具有重要意义。我们通过前期研究发现,在云南保山,海林芋果蝇在寄主植物爬树龙上进化出了卵丝的多型(这是昆虫卵期多型首次被发现):该虫将卵产于寄主花序内,其夏季世代卵丝短,一龄幼虫直接孵化取食佛焰苞;秋季世代(越冬世代)卵丝长,一龄幼虫在卵壳内于果序上越冬至翌年果实成熟后孵化,取食果肉。本项目基于寄主调查和卵形态比较推测其它几种芋果蝇是否转主寄生并采用卵多型策略,对确定为多型的物种,通过多因子实验确定其环境预示因子。果蝇卵丝是上皮图式和形态建成研究的重要模式系统,芋果蝇卵丝多型将为基于相同遗传背景解析图式和形态发生间的关联提供理想的研究模式。
英文摘要
Polymorphism is phenomenon that the same genotype produces two or more discrete phenotypes in response to environmental cues. Accurate prediction of future environment by factors acting as cues is critical for maintenance of polyphenism. Environmentally induced alterations in phenotypes of polyphenic organisms exhibit profoundly important functional, anatomical, and behavioral differences going far beyond superficial modifications of an otherwise genetically determined form. Ecological developmental studies with polyphenic organisms have important implications for the philosophy and practice of developmental biology. We found in our previous field investigation in Baoshan, Yunnan Province, that the drosophilid species, Colocasiomyia hailini, evolved a seasonally polyphenic egg filaments (dorsal appendages of egg shell) on its host plant, Rhaphidophora decursiva (this is for the first time that polyphenism during egg stage is found in insects): The female adults of C. hailini lays their eggs into the host inflorescences. Eggs of the summer generation(s) of this species are characterized by short filaments, and the 1st instar larvae hatched soon after oviposition and feed on the spathe of the host plant; however, eggs of the autumn (i.e., overwintering) generation of the same species are characterized by long filaments, with the 1st instar larvae dose not hatch until the dehiscence of the host infructescences – they then hatched and feed on mature pulp of the host. This project is proposed to determine whether several other Colocasiomyia species are heteroxenous and polyphenic in light of egg filaments, via filed investigation and morphological comparisons. For all species proved to be polyphenic in egg filaments, environmental cues triggering the developmental switch will be determined via multiple factor experiment. As Drosophila egg filaments (i.e., the dorsal eggshell structures) has emerged as an outstanding model for tubulogenesis, the egg filament polyphenims found in Colocasiomyia species are expected to provide an excellent system for studying the link between patterning and morphogenesis.
昆虫栖息于变化的环境中,为应对相关的生存压力,有的昆虫通过进化出特殊的表型可塑性即多型性来应对有规律地变化和可预期的环境,以维持自身的适合度。季节性多型是常见类型的多型,其允许昆虫在一年中的不同时期表达不同的表型,以适应时序上异质性的环境。这一紧密的环境-表型匹配的先决条件是存在一个可靠的环境预示信号,使多型昆虫能够基于该信号的诱导发育出在即将经历的环境中具有高适应度的表型。海林芋果蝇是一种来自双翅目果蝇科芋果蝇属的蝇类。该虫为适应其天南星科寄主植物——爬树龙Rhaphidophora decursiva而进化出的卵的多型性(卵丝相对卵体长度的间断变化以及卵体积的间断变化),并通过这一策略适应环境条件(温度、湿度和来自共寄主芋果蝇的竞争等)的季节性变化,使其种群在夏季得以连续多代“扩增”,并度过越冬休眠期间不利环境影响,极大地减少了与共寄主物种——长丝芋果蝇在产卵空间、幼虫期食物资源上的竞争。在本研究中,我们基于三因子实验对卵丝多型表型环境的诱导因子进行了推测。考虑到卵丝发育发生于母体卵巢内,环境预示信号极有可能是由母体所感知并通过跨代调控决定子代卵的发育,而由于卵巢的发育始于蛹期,我们优先考虑了海林芋果蝇自蛹期开始所经历的环境因子中,那些在夏、秋世代间变化显著且具有较好可预测性的三个环境因子——光周期、温度和共寄主芋果蝇(即长突芋果蝇)的存在与否,设计了三因子实验,基于对不同条件组合下雌成虫卵巢内卵子的表型(包括卵丝相对长度、卵轮廓外形、卵表面刻纹等)发育结果的统计比较分析,推测了这三个因子的变化对海林芋果蝇卵表型发育结果的影响。我们的研究结果表明,光周期是海林芋果蝇卵表型发育转换的主要环境诱导因子,温度对光周期的效应起到协同作用(高温对长日照有增效作用,低温则对短日照有增效作用),而长突芋果蝇的存在与否对卵表型的发育结果无显著影响。考虑到卵丝作为遗传和发育生物学中理解小管发生研究模式的重要性,我们的结果为探究环境因子在该过程中的作用及具体机制提供了机会。
