肝脏纤维化长期不断发展将导致其结构破坏和功能的丧失,目前其关键的分子机制仍不完全清楚。环状RNA在纤维化中的研究虽处初级阶段,但已有的研究已经有助于理解其在纤维化形成和发展中的价值。迄今为止尚未见环状RNA参与血吸虫感染导致肝脏纤维化形成的报道。本项目在已有研究基础上拟首先通过肝星状细胞高通量测序,q-PCR以及过表或消减等技术进一步验证环状RNA参与血吸虫感染肝脏纤维化的发生发展;然后采用Ago2-RIP、RNA pull down、双荧光素酶报告基因、RIP和EMSA等实验在细胞水平证明其可能通过竞争微小RNA作用或与蛋白相互作用的机制;再在血吸虫感染动物体内消减靶标分子后检测肝纤维化形成和发展相关因素;最后确定靶标circRNA同源的人源性circRNA存在于人肝脏纤维化标本上。关键环状RNA参与血吸虫肝脏纤维化形成和发展新机制的阐明,将为干预血吸虫肝脏纤维化的发生发展提供新的靶点。

The lasting development of liver fibrosis will eventually lead to the destruction of organ structure and loss of function. Although the research of circRNA in fibrosis is in the primary stage, the previous research has been helpful to understand the value of circRNA in the formation and development of fibrosis. circRNAs are thought to be related to the activation of stellate cells in liver fibrosis and may be a potential target to inhibit the development of liver fibrosis. Up to now, circRNAs have not been reported to be involved in liver fibrosis caused by schistosoma infection. On the basis of the our existing studies, this project intends to further verify the involvement of circRNAs in the occurrence and development of liver fibrosis caused by schistosoma infection through high-throughput sequencing, q-PCR, gene over-expression or gene knock down techniques. Then, the Ago2-RIP experiment, RNA pull down, double luciferase reporter gene, RIP, EMSA and other experiments were used to prove that it may compete for the action of microRNAs or interact with proteins at the level of hepatic stellate cells. The factors related to the formation and development of liver fibrosis were detected in schistosomiasis infected animals after knock down target molecules. Finally, it was determined that the homologous human circRNA of the target circRNA existed in the human liver fibrosis specimens. Elucidating the new mechanism of the involvement of key circRNAs in the formation and development of schistosomiasis liver fibrosis will provide a new target for the intervention and treatment of schistosomiasis liver fibrosis.
肝脏纤维化的形成和发展机制及干预靶点的研究一直为当今医学科学研究的热点和难点。本研究按项目计划先初步锚定了血吸虫感染小鼠肝星状细胞高通量测序发现的circGSR-0001、circFgfr2、CircKcnq1-0003和circAdpgk_0001等4个环状RNA,其均具有在6、9和12周不同感染时间差异表达最为显著的特性;完成了在血吸虫感染肝组织、原代星状细胞及活化的肝星状细胞株JS-1与测序结果一致的实验后确定其为靶标环状RNA。先验证了这4个靶标环状RNA符合环状RNA一般特点和在细胞胞质中定位;其次在细胞水平阐明了该4个靶标环状RNA具有对脏器纤维化形成有关的Col1α1和α-SMA的作用以及通过吸附抑制各自微小RNA(miR-383-3p、miR-1941-5p、miR-6354和miR-877-5p)的作用机制;实验还表明RNA结合蛋白FUS参与circFgfr2的环化形成。继而在血吸虫感染小鼠动物模型体内过表达或消减靶标环状RNA,结果均显示了可减少Col1α1和α-SMA的表达,减轻肝纤维化的程度,证明了这4个靶标环状RNA均参与了血吸虫肝脏纤维化形成和发展;最后,通过人体纤维化标本实验证实了circFgfr2和circAdpgk_0001与人肝脏纤维化(乙、丙肝病毒性、胆汁淤积性、非酒精代谢性、药物性和自身免疫性肝脏纤维化)具有显著的相关性且与纤维化严重程度密切相关。. 在项目执行过程中,发现高通量测序结果中部分高表达的环状RNA在星状细胞株采用小干扰RNA技术并不能消减其表达以鉴定功能。依据文献,我们建立了CRISPR/Cas13基因编辑技术成功消减了环状RNA circAdpgk_0001的表达并鉴定了其在星状细胞株的活化功能。. 本项目首次证实了关键靶标环状RNA参与血吸虫肝脏纤维化形成和发展并阐明了作用机制,不仅为血吸虫肝脏纤维化,也为其他病因导致的肝纤维化的发生发展提供了一系列新的干预靶点。
