The role of genes associated with cell growth in proliferating diseases

利用单克隆抗体和Northern blotting技术检测了银屑病组织中与细胞生长相关的几种原癌基因的表达，并与正常皮肤组织进行了比较。利用单克隆抗体rp35与ras p21反应的免疫荧光研究显示，在正常皮肤中，只有基础表皮层被染色，而在银屑病组织中，整个表皮层被染色，这表明该疾病中ras p21的过量产生。利用银屑病组织和正常皮肤的聚a(+) mRNA进行Northern印迹技术表明，与正常皮肤相比，银屑病组织中fos和myc转录本水平降低，而K-ras转录本水平升高。这些数据提示原癌基因与银屑病的发病机制有关。

Expressions of several protooncogenes related with cell growth were examined in psoriatic tissues comparing those in normal skin tissues using monoclonal antibody and Northern blotting techuiques. Immunofluorescence studies using monoclonal antibody rp35 reacted with ras p21 revealed that only based layers of epidermis were stained in normal skin, while whole layers of epidermis were stained in psoriatic tissues, sugges ting overproduction of ras p21 in this disease. Northern blotting techniques with poly a(+) mRNA derived from psoriatic tissues and normal skin demonstrated that levels of fos and myc transcripts were decreased, while that of K-ras transcript was increased in psoriatic tissues in comparison with those in normal skins. All these data suggest association of protooncogenes with pathogenesis of psoriasis.
