2-methacryloyloxyethyl phosphorylcholine(MPC) polymer is our original biocompatible polymer whose side chain is composed of phosphorylcholine that resembles phospholipids of biomembrane. The MPC hydrogel can be generated by mixing aqueous solutions of poly (MPC-co-methacrylic acid)(PMA) and poly(MPC-co-n-butyl methacrylate)(PMB). The present study investigated the effects of the MPC-hydrogel application on tendon adhesion and healing.1.Determination of the optimum condition for synthesis of MPC hydrogelVarious gels were generated by changing the molecular composition, molecular weight, structure of hydrophobic units, and evaluated in simulated body fluid and using rat model. The established optimal gel was used for the following studies 2-4.2. Investigation of the effects of MPC hydrogel for tendon healing..After a tendon was cut and sutured, the MPC hydrogel was applied to cover the injury site and the wound was closed.Tendon healing was evaluated by macroscopic, histological and mechanical testings.Macroscopic findings showed little or no adhesion was observed in the MPC group.HE staining showed that gaps of the injured tendon in both groups were similarly bridged by collagen fibers.There was no significant difference on tensile strength between two groups.3. Investigation of the effects of MPC hydrogel for prevention of tendon adhesion.After the same manner, tendon adhesion was evaluated by microscopic and mechanical testings.The findings showed severe adhesion in the control group. In contrast, little or no adhesion was observed in the MPC group.4. Evaluation of the lubrication property of MPC hydrogel covering a tendon suture site.By measuring of the tendon excursion resistance after cyclic flexion of the operated toe, the excellent lubrication property of MPC hydrogel was revealed.In conclusion, the MPC-hydrogel application immediately after the tendon suture may prevent the adhesion without impairing the healing.
