This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Our recent animal studies have demonstrated that administration of sesame oil to atherosclerotic mice dramatically reduced serum lipids, inflammatory markers, and overall decreased atherosclerotic process. Sesame oil is a non-toxic edible oil, which is rich in both mono-unsaturated and poly-unsaturated fatty acids, plus it contains lignans and antioxidant (non-saponifiable) components. Numerous health benefits have been attributed to the oil. Although, pharmacological agents are available for the reduction of serum lipids, they are expensive and have side effects with prolonged use. Therefore, it is essential to describe whether sesame oil would have beneficial effects on markers of atherosclerosis in humans. This study will determine whether supplementation with sesame oil will provide cardiovascular benefits as measured by changes in lipid/lipoprotein levels, glucose homeostasis, inflammatory and oxidative biomarkers, beyond what is seen in oil with analogous fatty acid composition. The study will use normal subjects and should provide valuable insights into the potential importance of non-saponifiable components of sesame oil. We propose the following specific aims to determine the efficacy and safety of sesame oil consumption: 1) To determine the effect of sesame oil on markers of cardiovascular risk in normal, healthy subjects. Specifically, we will determine any effect, after sesame oil supplementation, on the lipid lipoprotein profile, blood pressure, glucose homeostasis, inflammatory and oxidative stress biomarkers. 2) To determine whether the non-saponifiable components of sesame oil are modulating cardiovascular risk by comparing above mentioned results with the endpoints from normal, healthy subjects who consume an oil mixture of analogous fatty acid composition.
