童年期不良经历对青少年自杀行为的影响：基因-脑-行为的多角度研究

青少年自杀是一个严重的社会和公共卫生问题。课题组前期研究发现，童年期不良经历（ACE）可致青少年自杀风险增加，但其中的生物学机制尚不明确。已有研究证实表观遗传改变是个体适应环境压力的重要机制，那么ACE可否通过改变DNA甲基化影响青少年的自杀行为？且相关的脑功能改变是否在DNA甲基化与自杀行为间起中介作用？目前尚不清楚。本研究拟从基因-脑-行为的系统研究思路出发，以SKA2基因作为突破口，利用基因分析和脑功能成像等技术手段，结合横断面调查，从表观遗传、神经系统和行为心理等不同层面研究SKA2基因甲基化、情绪调节神经环路及心理特征在ACE影响青少年自杀行为中的作用，以期初步揭示ACE致青少年自杀行为发生的生物学机制及阐明其中的心理机制，为青少年自杀行为的预防提供理论依据以及为干预提供新靶点。

Suicide in adolescents is a serious social and public health problem. Our previous study found that adverse childhood experiences (ACE) increase the risk of suicide in adolescents, but the inside biological mechanism is still unclear. The existing studies have confirmed that epigenetics is an important mechanism for individuals to adapt to environmental stress, then can ACE influence suicidal behavior among adolescents by changing DNA methylation? Do related changes in brain function mediate the relationship between DNA methylation and suicidal behavior? It's not clear. The present study guided by gene - brain - behavior framework, take SKA2 gene as the breakthrough point, and combine cross-sectional investigation with gene analysis and functional MRI, explore the role of SKA2 gene methylation, neural circuits of emotion regulation and psychological characteristics in ACE affect suicide among adolescents from the aspect of epigenetics, nervous system and psychology, so as to preliminarily reveal the biological mechanisms and clarify the inside psychological mechanism of ACE on suicide behavior in adolescents, provide theoretical basis for the prevention and new targets for intervention of youth suicide.
