Both stress and alcohol exposure can lead to neurological disorders. Their impact on brain regions like the central amygdala (CeA) may underly the high prevalence of stress-related and alcohol use disorder comorbidities. The kappa opioid receptor (KOR) system is engaged by both stress and chronic ethanol exposure, and there is increasing evidence that together with its endogenous ligand, dynorphin (DYN), the KOR system is critically involved in the manifestation of these disorders (Figure 1). However, the role of the DYN/KOR system in these disorders is often studied independently. While these types of studies are important and informative, they leave a large gap in the preclinical field of psychiatric disorders. To adequately treat patients with comorbid stress-related and alcohol use disorders, more research must be done to understand the interaction and influence stress and alcohol use has on one another. Haun et al.(1), in the current issue of Biological Psychiatry, highlight the need for research focused on comorbidities. Using messenger RNA (mRNA) measurements, chemogenetics, and pharmacology, the authors provide a convincing argument for a critical role of the CeA DYN/KOR system in mediating stress-induced increases in alcohol consumption.To determine the influence of stress on alcohol intake, the authors use a novel and robust model of stress-enhanced alcohol drinking that incorporates chronic intermittent alcohol exposure and forced swim stress (CIE+ FSS).
