Dr Springer has assembled a collection of articles by distinguished authors which covers most aspects of the production and use of monoclonal antibodies (mAb). it is not primarily a 'cookbook' of experimental protocols, although a few chapters do contain detailed methods, but a series of minireviews which explain the literature and show how the technology can be used. There are five main sections. Part I entitled 'The production and improvement through genetic en- gineering of monoclonal antibodies' covers both mouse and human hy- bridoma production. It also contains information on how to obtain variants of the antibody first pro- duced if it isn't entirely satisfactory. To me, however, the most interest- ing chapter was that by AIt on the expression of cloned immunoglobu- lin genes. Part II covers some of the many applications of monoclonal anti- bodies to biomedical research. Many aspects of cell surface receptor stud- ies are covered as well as the de- lineation of antigenic determinants on viruses and the receptors for viruses on cells. There is a valuable chapter by Parham covering tech- niques for producing fragments of monoclonals as well as the binding of antibodies to cell surfaces. Part III outlines some of the tech- niques used to clone and transfect genes. It is particularly strong on the use of antibodies either to purify messenger RNA for specific proteins and to detect expression of the gene product after cloning. Part IV contains four chapters on applications in diagnosis and ther- apy. This is the most limited section of the book. It covers only the use of mAb to clear bone marrow of un- wanted cells using complement fix- ing -antibodies, the therapy of organ grafts and leukemias with mAb, and a very short outline of their potential use to remove toxic drugs or to visualize specific pathological pro- cesses such as infarcts or thrombi. Anyone seeking a fuller discussion of the clinical uses of mAbs should consult McMichael and Fabre's Monoclonal Antibodies in Clinical Medicine which is the closest com- petitor for this volume. The final part of this book de- scribes some of the uses of T-cel! hybridomas and T-cell clones as sources of biologically active prod- ucts. An overview of their use in defining the T-cell antigen receptor and the study of the cytolytic process is included. In summary, this book provides background information on a num- ber of topics in rapidly evolving areas of biotechnology. As such it would be of value to many graduate stu- dents in the biosciences and phys- ician investigators interested in understanding the ways in which antibodies can be used to solve problems. It is not, and is not in- tended to be, a catalogue of detailed methodology or a comprehensive list of clinical applications. I enjoyed reading it and I learned several things from it.
